Objective In this study,under the guidance of the clinical experience of Professor Zhu Zongyuan,a well-known TCM doctor in Inner Mongolia Autonomous Region,in the treatment of chronic atrophic gastritis(Chronic Atrophic Gastritis,CAG),the effect of his experience prescription Modified Huangqi Jianzhong Decoction on prevention and treatment of CAG rats was observed.The pathological changes and the expression of related factors explain the possible mechanism of action of the prescription,and provide ideas and theoretical basis for the clinical promotion and application of Chinese medicine.Methods Forty-eight SD rats were selected.After one week of adaptive feeding,they were given 180 μ g/ml MNNG free drink combined with 40% ethanol twice a week by gavage to replicate the CAG rat model.After successful modeling,they were randomly divided into normal group,model group,vitamin enzyme control group,and low,medium and high dose groups of Modified Huangqi Jianzhong Decoction,with 8 rats in each group.After the model was successfully established,intragastric administration was started.After12 weeks of drug intervention,the general condition and weight changes of the rats were observed.The pathological changes of gastric mucosa were observed by HE staining.The apoptosis rate of gastric mucosal epithelial cells was detected by flow cytometry,Immunohistochemical method was used to detect the expression of Ki67,caspase3,Cyto-C and cell proliferation and apoptosis related factors.Result 1.Analysis of the body weight of rats in each group: the weight level of rats in the model group was lower than that in the normal group(P<0.05);while the weight levels of the rats in the vitamin enzyme control group and the Modified Huangqi Jianzhong Decoction low,medium and high groups were equal Higher than the model group(P<0.05 or P<0.01),the body weight of rats in the low,medium and high groups of Modified Huangqi Jianzhong Decoction was significantly higher(P<0.01).2.Pathological observation of gastric mucosal tissue under light microscope: Compared with the normal group,each group had gastric mucosal thinning,irregular edges,necrosis of mucosal epithelium and proper glands,edema of gastric glandular gland cells,and massive inflammation in each group after modeling.The pathological changes of cell invasion;compared with the model group,the arrangement of gastric glands in each treatment group tended to be neat,the number of inflammatory cells decreased,and gastric mucosal atrophy was improved to varying degrees;the medium and high-dose groups of Modified Huangqi Jianzhong Decoction improved the mucosa The effect of adjusting the arrangement of glands,anti-inflammatory and reducing glandular cell edema is better than that of the control group.3.Ki67 expression in gastric mucosal tissue of rats in each group: Ki67 expression in the model group was significantly lower than that in the normal group(P<0.01);while the expression of Ki67 in the medium and high doses of Modified Huangqi Jianzhong Decoction was significantly higher than that of the model group(P<0.01);Comparing the two different drug intervention groups,the expression of Ki67 in the medium and high-dose groups of Modified Huangqi Jianzhong Decoction was significantly higher than that of the vitamin enzyme control group(P<0.01);each group of the Modified Huangqi Jianzhong Decoction group In comparison,the Ki67 expression in the low-dose group and the high-dose group of Modified Huangqi Jianzhong Decoction were significantly higher(P<0.01).4.Expression of cell apoptosis rate in gastric mucosa of rats in each group: The cell apoptosis rate in the model group was significantly higher than that in the normal group(P<0.01);the vitamin enzyme control group and the low-dose group of Modified Huangqi Jianzhong Decoction were compared with the model The apoptotic rate of the group was low(P<0.05);the apoptosis rate of the medium and high-dose groups of Modified Huangqi Jianzhong Decoction was significantly lower than that of the model group(P<0.01).5.The expression of caspase3 in gastric mucosa of rats in each group: the expression of caspase3 in the model group was significantly higher than that in the normal group(P<0.01);while the expression of caspase3 in the treatment group was lower than that in the model group(P<0.05 or P< 0.01),with Modified Huangqi Jianzhong Decoction significantly better;comparison between the treatment groups,the expression of caspase3 in the high-dose Modified Huangqi Jianzhong Decoction group was lower than that of the vitamin enzyme control group(P<0.05);each group of Modified Huangqi Jianzhong Decoction Compared with the low-dose group of Modified Huangqi Jianzhong Decoction,the expression of caspase3 in the high-dose group of Modified Huangqi Jianzhong Decoction was decreased(P<0.05).6.The expression of Cyto-C in gastric mucosal tissues of rats in each group: The expression of Cyto-C in the Vimein control group,the medium and high-dose groups of Modified Huangqi Jianzhong Decoction was lower than that of the model group(P<0.05 or P< 0.01),the medium and high doses of Modified Huangqi Jianzhong Decoction were superior;compared with the control group,the expression of Cyto-C in the medium dose group of Modified Huangqi Jianzhong Decoction decreased(P<0.05),and the expression of Cyto-C in the medium dose group of Modified Huangqi Jianzhong Decoction was lower(P<0.05).The expression of Cyto-C in the high-dose group was significantly reduced(P<0.01);compared between the groups of Modified Huangqi Jianzhong Decoction,the expression of Cyto-C in the medium and high-dose groups of Modified Huangqi Jianzhong Decoction was significantly reduced in the low-dose group(P<0.01).Conclusion The well-known medical prescription Modified Huangqi Jianzhong Decoction has a good preventive effect on chronic atrophic gastritis;the mechanism of this prescription may be related to the regulation of the expression of Ki67,caspase3,Cyto-C,improve the morphological changes of gastric mucosa tissue,and regulate gastric mucosal cells The unbalanced state of proliferation and apoptosis,and the specific ways of action need further study.
