Saffron Aqueous Extract Component Analysis And Depression Pharmacological Analysis

Nowadays,depression has become one of the most common mental diseases,but the research on depression drugs is lagging behind.Saffron as a traditional Chinese medicine,the main effect is to promote blood circulation and remove blood stasis,tranquilize and relieve depression.Some Chinese doctors think it can be used to treat depression.This study aims to test whether saffron has antidepressant effect and explore the possible mechanism,so as to provide sufficient experimental basis for further research and development of saffron.In this study,the best extraction method of saffron was explored by using different solid-liquid ratio,different extraction time,different temperature,grinding and ultrasonic extraction,and the components of Crocus sativus water extract were analyzed by liquid chromatography tandem mass spectrometry.The effect of saffron on depression in mice was detected by comparing the social interaction experiment and serum glucocorticoid content between the depressed mice and the depressed mice with intraperitoneal injecting of saffron.The effects of saffron aqueous extract on neural stem cells in hippocampus of depression mice were detected by immunohistochemistry with Ki67 and DCX as markers to evaluate whether saffron has antidepressant effect by increasing hippocampal neurogenesis.The results showed that crocin and picrocrocin were the main components in saffron water extract.The results of social experiment showed that saffrom water extract could significantly increase the contact time between depressed mice and healthy mice.The results of glucocorticoid test showed that saffron water extract could reduce the content level of glucocorticoid in serum of depressed mice.The results of immunohistochemistry showed that saffron water extract could significantly increase the contact time between depressed mice and heathy mice.The results showed that the number of DCX positive cells and the number of DCX positive cells were significantly increased.In conclusion,saffron water extract can not only reduce depression like behavior and glucocorticoid levels,but also increase neurogenesis in depression mice.This study revealed that has high comprehensive utilization value.saffron water extract may reduce the depression like behavior of depression mice by improving the serum glucocorticoid level and increasing the neurogenesis in the DG area of hippocampus.This work provides an experimental basis for the role of saffron in the treatment of depression.
