| Objective:The effects of electroacupuncture scalp point-through-point therapy on motor function recovery of MCAO model rats were observed dynamically,and the differences in motor function and the expression of Fibulin-5 in the right cerebral hemisphere of rats were compared and analyzed.To investigate the mechanism of electroacupuncture on the head to improve motor dysfunction in rats with cerebral infarctionMethods:made middle cerebral artery occlusion(MCAO)mode by the method of suture-occluded method.Male Sprague Dawley rats meeting the requirements were randomly divided into blank group,sham operation group,model group and electroacupuncture group(2Hz,50 Hz and 100Hz),with 10 rats in each group.Among them,the blank group and the model group were treated with the same bundle method as the electroacupuncture group,the time was20min;In the electroacupuncture group,Baihui and Qianshencong acupoint was selected and stimulated by continuous wave of 2Hz,50 Hz and 100 Hz respectively.The treatment lasted for 20 min each time,once a day,for a total of 14 days.On the 1st,3rd,5th,7th and 14 th day of electroacupuncture treatment,the motor function of the six groups of rats was measured by the balance beam test.The positive expression of fibulin-5 in the right cerebral hemisphere of rats was observed by immunohistochemical(IHC).The expression of Fibulin-5 in the right cerebral hemisphere of rats infarction area was quantitatively analyzed by enzyme-linked immunosorbent assay(ELISA).SPSS 23.0 statistical software was used to analyze the experimental data.Results:1.Effects of different frequencies of electroacupuncture on motor function rehabilitation of rats with cerebral infarctionThere was no significant difference in the behavior scores between the sham operation group and the blank group(p > 0.05).The removal of tissues and blood vessels during the preparation of the MCAO model by thread embolization had no effect on the motor function of the rats;The difference of behavioral scores between model group and blank group was significant(p <0.01).The motor function of MCAO model rats prepared by thread embolization method could be reduced;The behavioral scores of MCAO model rats in each electroacupuncture group increased gradually with time,and on the 14 th day,the scores were close to the blank group,and the difference was statistically significant compared with the model group(p < 0.01).The 2Hz electroacupuncture group was significantly different from the model group after the 3rd day of treatment(p< 0.05),and significantly different from the50 Hz group and the 100 Hz group after the end of treatment(p < 0.05).The50 Hz and 100 Hz electroacupuncture groups were significantly different from the model group after the 5th day of treatment(p < 0.05),and there was no significant difference in efficacy between the two groups after the end of treatment(p > 0.05).It is suggested that electroacupuncture can promote the recovery of motor function in MCAO model rats,and the effect of 2Hz electroacupuncture group is the best.2.Qualitative and quantitative analysis of fibulin-5 expression in brain tissue of rats in each group:Immunohistochemical assay showed that the expression of fibulin-5 was plasmically positive in the right cerebral hemisphere,mainly distributed in the cytoplasm or cytoplasmic matrix.Immunohistochemistry and ELISA quantitative analysis showed that:After 14 days of electroacupuncture on the head,the expression of Fibulin-5 in the right cerebral hemisphere of rats in the model group and each electroacupuncture group was significantly increased compared with the blank group,but the expression promotion effect of electroacupuncture groups with different frequencies was different,among which the 2Hz group was the most significant and the difference was statistically significant compared with the 50 Hz group and the 100 Hz group(p < 0.05).There was no significant difference between the 50 Hz group and the 100 Hz group(p> 0.05).3.Correlation analysis of behavioristics-beam balance test score and the expression of fibulin-5 in ischemic brain tissue in rats:Behavioral evaluation of MCAO model rats in the 2Hz electroacupuncture group--the correlation coefficient between the balance beam walking test score and the expression of Fibulin-5 in ischemic brain tissue was 0.76 and p < 0.05,indicating that the motor function recovery of rats with cerebral infarction may be related to the expression of Fibulin-5.Conclusion:1.Head electroacupuncture can effectively promote the recovery of motor function in MCAO model rats,and different frequencies of electroacupuncture have different effects on the improvement of motor function recovery,among which 2Hz electroacupuncture group has the best effect.2.Head electroacupuncture can promote the expression of Fibulin-5 in the ischemic penumbra of MCAO model rats,and different frequencies have different effects,among which the 2Hz electroacupuncture group has the best effect of promoting the expression.3.Head electroacupuncture can effectively protect the brain tissue of rats with cerebral infarction and promote the recovery of motor function of rats with cerebral infarction.The mechanism may be to up-regulate the expression of Fibulin-5 in ischemic brain tissue,and then regulate the function of extracellular matrix to protect neurovascular units and promote rehabilitation. |
