Objective: To observe the changes of intestinal flora of patients with Yang-deficiency syndrome after chemotherapy in acute leukemia(AL),and to explore the relationship between the occurrence of Yang-deficiency syndrome and changes in flora after acute leukemia(AL)chemotherapy.The effect of Shenfu injection on the intestinal mucosal barrier of mice with Yang-deficiency after chemotherapy was explored through animal experiments.Summarize the characteristics of the intestinal microenvironment of Yang Deficiency Syndrome and guide clinical medication.method:1.Collect 15 clinical cases of AL patients with Yang-deficiency syndrome after standard chemotherapy,and collect the general data of the patients.The stools of patients before chemotherapy,2 weeks after administration,and 3 weeks after administration were collected for 16 S r RNA detection.Analyze the correlation between Yang-deficiency syndrome,changes in intestinal flora and adverse reactions after chemotherapy.2.Using 20 Balb/C male mice,5-fluorouracil(50mg/kg)was injected intraperitoneally for 5 consecutive days,combined with Shenfu injection(2ml/kg;4ml/kg)for 5 consecutive intraperitoneal injections,the sixth day Blood was collected from the tail vein,and the mice were sacrificed on the 8th day.The effect of Shenfu injection on the intestinal mucosal barrier of mice with yang deficiency after chemotherapy was observed by HE staining,routine blood test,and fluorescence test.result:1.The effect of yang deficiency syndrome on the intestinal flora of patients with acute leukemia after chemotherapyIn the collected 15 patients with yang deficiency syndrome after AL chemotherapy,the diversity of intestinal flora decreased significantly in the second week after chemotherapy compared with before chemotherapy(P=0.0246).In the 3rd week after chemotherapy,the main bacteria of the intestinal flora changed significantly compared with that before chemotherapy,and the main bacteria were pathogenic bacteria.Antibiotics have a significant effect on the diversity of intestinal flora in patients with Yang deficiency after chemotherapy(P=0.046).The occurrence of fever and diarrhea in patients with Yang deficiency during chemotherapy may be related to the decrease of flora diversity and the recovery of blood picture after chemotherapy,but no significant correlation has been found(P>0.05).2.The effect of Shenfu injection on the intestinal mucosal barrier of Yang-deficiency mice after 5-fluorouracil chemotherapyShenfu injection can protect the intestinal mucosal barrier by reducing the damage of chemotherapy to intestinal tissues,and significantly reduce the change of intestinal permeability compared with the model group(P=0.0229).Shenfu injection can also significantly delay the weight loss of Yang-deficiency mice and the occurrence of diarrhea after chemotherapy,and at the same time improve the blood picture(P<0.05).Conclusion: After chemotherapy,the Yang Qi level of Yang Deficiency Syndrome is significantly related to the decrease of intestinal flora diversity and the proliferation of pathogenic bacteria.Shenfu injection can improve the survival status of mice with Yang deficiency syndrome after chemotherapy by protecting the intestinal mucosal barrier.It is suggested that Shenfu injection and other traditional Chinese medicines can be used to treat patients with yang deficiency after chemotherapy for acute leukemia.The main mechanism is to improve the intestinal microenvironment,which is specifically manifested in adjusting the diversity of intestinal flora and protecting the intestinal mucosal barrier.
