Research objectives:To observe the effects of electroacupuncture at "Shuigou" acupoint on cerebral vascular calcitonin gene-related peptide(CGRP),arginine vasopressin(AVP)and angiotensin Ⅱ(ANG-A)in rats with focal cerebral ischemia,and to explore the mechanism of electroacupuncture at "Shuigou" acupoint in the treatment of ischemic cerebrovascular diseases.The research methods:According to the random number table method,30 healthy male Wistar rats meeting the inclusion criteria were randomly divided into electroacupuncture group,model group and sham operation group with 10 rats in each group.Meanwhile,the electroacupuncture group,model group and sham operation group were divided into 3 d and 7 d according to the duration of treatment,with 5 rats in each group at each phase.MCAO model rats with middle cerebral artery occlusion(MCAO)were prepared by electroacupuncture group and model group using thread embolization method.The rats in the sham operation group and model group did not receive other intervention treatment,but the binding time was the same as that in the electroacupuncture group.Electroacupuncture group 3d acupuncture "Shuigou" point in the middle of the cleft lip and 1 mm below the nose tip of rats.A 0.35 mm × 13 mm needle was used to obserously prick 0.2 ~ 0.3cm from Shuigou point to the direction of nasal septium,and then Hans-100 A Han electroacupuncture treatment instrument was connected.The negative pole was connected to "Shuigou" point,and the positive pole was connected to 2 mm below "Shuigou" point.Continuous wave,frequency 15 Hz,intensity 1 m A,electroacupuncture treatment for 20 min,once a day,consecutive 3 days.Electroacupuncture group for 7 days: The operation was the same as electroacupuncture group for 3 days,and the treatment was continuous for 7 days.Inclusion and exclusion criteria were based on Zea Longa method.Neurological symptoms of each group were scored immediately after surgery,3 days after surgery,and 7 days after surgery,respectively.Neurological impairment of the rats was evaluated by referring to the Modified Neurological Severity Score(NSS).Cerebrovascular tissues of the anterior cerebral artery,middle cerebral artery and posterior cerebral artery of rats in each group were separated and extracted on the 3rd and 7th day after operation.The levels of calcitonin gene-related peptide(CGRP),arginine vasopressin(AVP)and angiotensin Ⅱ(Ang-Pin)in cerebral vascular tissues of cerebral vessels on both sides of the hemisphere in infarcted and non-infarcted MCAO rats in each group were detected by ELISA.SPSS21.0 software was used to create a database for all experimental data,and statistical analysis was performed.Results:1.The results of NSS score of rats in each group showed that there was no statistical difference in the neurological deficit between the electroacupuncture group and the model group immediately after surgery,indicating that there was no significant difference in the neurological deficit between the electroacupuncture group and the model group before electroacupuncture intervention.After modeling,the NSS score in model group was significantly higher than that in sham operation group(P < 0.01),and the difference was statistically significant.Compared with model group,NSS score of electroacupuncture group was significantly decreased(P < 0.01),and the difference was statistically significant.Compared with the electroacupuncture group,the NSS score of MCAO rats decreased significantly after electroacupuncture 3 d,and significantly decreased in the electroacupuncture group 7 d(P < 0.01).The difference was statistically significant.2.The contents of CGRP,AVP and Ang-Ⅱ in bilateral cerebral vascular tissues of rats in the infarcted hemispheres and non-infarcted hemispheres in each group showed as follows:compared with the sham operation group,the contents of CGRP,AVP and Ang-mol in the cerebral vascular tissues of rats in the infarcted hemispheres in the model group were significantly increased(P < 0.05),with statistical significance.Compared with model group,the content of CGRP in cerebral vascular tissue of infarction hemisphere in electroacupuncture group for 7 d was significantly increased,while the content of AVP and Ang-Ⅱ was significantly decreased(P < 0.01),with statistical significance.The levels of CGRP in cerebral vascular tissues of both the infarcted and non-infarcted hemispheres were increased in the electroacupuncture group for 7 days,especially in the infarcted hemispheres(P < 0.05).The contents of AVP and Ang-Ⅱ in cerebral vascular tissue of infarction hemisphere in electroacupuncture group for 7 d were significantly decreased(P < 0.05),and the difference was statistically significant.The contents of AVP and Ang-Ⅱ in cerebral vascular tissue of the infarction hemisphere were decreased at 3d and 7d after electroacupuncture intervention,and the contents of AVP and Ang-bar were significantly decreased at 7d after electroacupuncture intervention(P < 0.05),the difference was statistically significant.The content of CGRP in cerebral vascular tissue of the infarct hemisphere increased at 3 d and 7 d after electroacupuncture,but there was no significant difference(P > 0.05).Conclusions:1.After the occurrence of cerebral ischemia,the motor function of rats was significantly impaired,and the NSS score was significantly increased.After electroacupuncture,the NSS score was significantly decreased,and the motor function of rats was significantly improved.2.The levels of AVP and Ang-Ⅱ in cerebral vascular tissue of the infarct hemisphere were significantly increased,while the levels of CGRP were significantly decreased,suggesting that CGRP,AVP and Ang-P may be involved in the pathological process of ischemic brain injury.3.Compared with the model group,the content of CGRP in cerebral vascular tissue of cerebral infarction hemisphere in electroacupuncture group was significantly increased,while the content of AVP and Ang-Ⅱ was significantly decreased,and with the extension of intervention time,the content of AVP and Ang-protein showed a trend of gradual decline.It is suggested that the benign regulation of CGRP,AVP and Ang-Ⅱ in ischemic cerebrovascular tissue may be one of the mechanisms of electroacupuncture in the treatment of cerebral infarction. |
