Objective To establish a simple,reliable and minor trauma model of focal cerebral ischemia in wistar rats.Methods The focal cerebral ischemia model in wistar rats were made by the modified suture-occuluded method of Zealonga and Koizum.Then the reliability of this model was evaluated with the examination of nearoethohgy.Results 2 hours after MCAO/R rats showed neurologincal functional defecf.Conclusion The improved model is simple,reliable,minor traumatic and stable.It is advisable to use this experimental animal model to study focal cerebral ischemia.Obective To investigate the dynamic changes of neuroglobin expression in rats following middle ischemic/reperfusion injury.Methods The middle cerebral artery occlusion(MCAO) (for 2 hours)/reperfusion(R) model in rats(MCAO/R group),was established by the modified suture-occluded method of Zea Longa.Then they were randomly divided into 8 groups according to the seven time points of 1h,4h,8h,16h,32h,64h and 128h afier ischemia/reper-fusion and sham group.We used the methods of nerve function evaluation,detection with light microscope to evaluate the MCAO/R rat models at different time points,analyzing the changes of the expression of NGB in the center and the ischemic penumbra at different time points by immunohistochemical methods coupled with computer - assisted image analysis.Results The changes in NGB expression were different in cerebral cortex and in the hippocampus.NGB pro- tein was regulated from rising to declining ischemia/reperfusion insult to the periphery cerebral cortex,but its positive expression did not change over time in the ipsilateral cortical symmetric region,while it keeps always down - regulated tendency in the hippocampus.Pathology showed the degree of hippocampus nerve cell's tranma was greater than the that of cerebral cortex.Conclusions That different endurance to ischemic damage to different area in the brain may be in relation to the expression of endogenous NGB in the brain following ischemia/ reperfusion.The increased expression of endogenous NGB in the brain after ischemia/reperfusion may be associated with the protective response to ischemic damage.
