1 PurposeBased on the corresponding theory of heaven and man of traditional Chinese medicine,proved that the allergic immune response of college students had seasonal rhythmicity,and explored the rhythmicity of SLFN12 gene expression level in peripheral blood of college students under different solar terms,and revealed that there was a correlation between the seasonal incidence of allergic rhinitis and the changes of SLFN12 gene mRNA and protein levels,in order to provide some clinical reference for the prevention and treatment of allergic rhinitis.2 MethodAccording to the inclusion and exclusion criteria,40 cases of each solar terms in the allergic group and the healthy group(20 cases in the allergic group,20 cases in the healthy group,10 cases in each male and female),and 6 solar terms were repeatedly measured(Qing Ming,Xiao Man,Xiao Shu,Chu Shu,Han Lu,Xiao Han).ELISA in human serum of different solar terms test detection of IgE,HIS,IL-17and LTC4.CELLAVISIONDM96 blood cells in nasal mucosa of different solar terms test morphological analysis of EOS and LYM.Western Blotting method and Real-Time PCR method test levels of SLFN12gene mRNA and protein in peripheral blood of people with different solar terms.Statistical analysis using SPSS23.0 software,the mean±standard deviation represents statistical data,and the research method is two-factor repeated measurement analysis of variance.The correlation among the indicators was analyzed.3 Results3.1Effects of different solar terms on the content of allergic and immune-related substances in human serumSerum IgE:In the allergic group,the expression was high in Xiao Man and low in Qing Ming.In the healthy group,the expression was high in Han Lu and low in Xiao Shu.Serum HIS:The allergic group had high expression in Xiao Man and low expression in Han Lu.While high expression in Xiao Han and low expression in Xiao Shu in the healthy group.Serum IL-17:In the allergic group,the expression was high in Xiao Han and low in Xiao Shu.In the healthy group,the expression was high in Qing Ming and low in Xiao Shu.Serum LTC4:Both in the allergic and healthy group,the expression was high in Qing Ming and low in Xiao Shu.3.2 Effects of different solar terms on the contents of EOS and LYM in human nasal mucosal secretionsEOS score in nasal mucosal secretions:The allergic group showed high expression in Qing Ming and low expression in Xiao Shu.In healthy group,high expression in Han Lu and low expression in Xiao Shu.LYM score in nasal mucosal secretions:The allergic group showed high expression in Qing Ming and low expression in Xiao Shu.In healthy group,high expression in Qing Ming and low expression in Chu Shu.3.3 Effects of different Solar terms on the level of SLFN12 Gene in Human Peripheral BloodResults of SLFN12 gene at mRNA level:In allergic group,the expression was high in Qing Ming and low in Xiao Man.In the healthy group,the expression was high in Han Lu and low in Xiao Shu.Results of protein expression of SLFN12 gene:The allergic group had high expression in Han Lu and low expression in Xiao Man.While high expression in Han Lu and low expression in Xiao Man in healthy group.3.4 Study on the relationship between SLFN12 gene level and allergic immune related substances in peripheral blood of different solar termsmRNA level:In allergic group:Chu Shu and IgE,HIS have negative correlation;In healthy group:Qing Ming and HIS have positive correlation.Protein level:In allergic group:Qing Ming and Han Lu had negative correlation with EOS,Han Lu had negative correlation with LYM;In healthy group:Qing Ming had negative correlation with IL-17,Xiao Han had positive correlation with IgE,HIS and IL-17.4 Conclusion(1)The state of allergic immune response of human body has seasonal rhythm,which is characterized by the high expression of Qing Ming Festival and Han Lu solar terms,that is,the allergic immune state of human body is hyperactive in spring and autumn.(2)The change of SLFN12 gene is related to the pathogenesis of allergic rhinitis,and has the characteristics of seasonal variation,which is characterized by the high expression of Han Lu solar terms,that is,it is closely related to the pathogenesis of allergic rhinitis in autumn.(3)The change of SLFN12 gene is correlated with the change of allergic immune response,and the change trend is basically the same.
