| Indigestion Tab.t is a traditional Chinese patent medicine that is often used in the homes of the majority of people.Radix Pseudostellariae has the function of nourishing the spleen and lungs,replenishing qi and promoting fluid,and is one of the main raw materials of Indigestion Tab.ts.The first part of the 2020 edition of the Chinese Pharmacopoeia only sets requirements for five items: traits,identification,moisture,total ash,and extracts of Radix Pseudostellariae,and cannot evaluate the quality and differences of medicinal materials produced in different regions.In this project,UPLC-LTQ-Orbitrap technology will be used to analyze the metabonomic differences of Radix Pseudostellariae from the 5 origins of Anhui,Guizhou,Hebei,Shandong,and Fujian,and explain the characteristics of the metabolites and metabolic pathways of Radix Pseudostellariae from different origins,in order to further improve the quality of Radix Pseudostellariae.Evaluation and selection of high-quality and stable Radix Pseudostellariae provide scientific and reasonable reference basis.The research content of this topic is mainly composed of the following three parts:(1)By consulting the literature,using UPLC-LTQ-Orbitrap analysis technology to collect the mass spectrometry information of the metabolites of Radix Pseudostellariae through the high-resolution mass-to-charge ratio,human metabolome database,massbank database combined with the secondary cleavage law of the compound,the metabolites of Radix Pseudostellariae were identified.A total of 100 kinds of metabolites were identified,basically including amino acids,peptides,nucleosides,etc.,which are common metabolites of Radix Pseudostellariae,providing a reasonable basis for metabonomic analysis of Radix Pseudostellariae samples.(2)Import the collected mass spectrum data into Thermo sieve software for denoising,peak alignment,peak extraction and other data preprocessing to obtain the data matrix of metabolites;and further import the matrix data into SIMCA-P 14.1(Umetrics,ume(?),Sweden)software,The data is normalized and standardized,and the PCA analysis,OPLS-DA analysis,correlation analysis between metabolites and hierarchical clustering analysis are performed.The results show that there are significant differences in metabolites of Radix Pseudostellariae from different origins,and are based on amino acids and peptides.,Nucleosides are the most important,and amino acids,peptides,and nucleosides are also one of the main active components of Radix Pseudostellariae.(3)Combined with databases such as KEGG to analyze the metabolic pathways of Radix Pseudostellariae from different origins,the results show that there are significant differences in the metabolic pathways of Radix Pseudostellariae from different origins.A total of 21 metabolic pathways are involved,namely aminoacyl biosynthesis,valine,and leucine.Degradation metabolism of acid and isoleucine,biosynthesis of valine,leucine and isoleucine,purine metabolism,alanine,aspartic acid and glutamic acid metabolism,arginine biosynthesis,glyoxylic acid and dicarboxylic acid metabolism,nitrogen metabolism,pyrimidine metabolism,zeatin biosynthesis,arginine and proline metabolism,glutathione metabolism,methyl butyrate metabolism,porphyrin and chlorophyll metabolism,pantothenate and coenzyme a biosynthesis,tryptophan metabolism,glycerophospholipid metabolism,glycine,serine and threonine metabolism,fatty acid biosynthesis,α-linolenic acid and linoleic acid metabolism,phospholipid biosynthesis.Through the analysis of metabolic pathways in Radix Pseudostellariae,there are significant differences in amino acid,peptide synthesis and metabolic pathways in Radix Pseudostellariae from different origins.A total of 8metabolic pathways are involved,indicating that the geographical environment is more sensitive to the synthesis and metabolism of amino acids and peptides in Radix Pseudostellariae. |
