| This research was to investigate the therapeutic and interventional effect of RPSLP(The stems and leaves of Radix pseudostellariae)on DSS(Dextran Sulfate Sodium)induced ulcerative colitis mice via valuating the efficacy of the anti-inflammatory and protection of colon epithelia.It will provide a new evidence for clinical application of RPSLP.1.To evaluate the efficacy and homogenicity of two acute colitis models induced by drinking 3%DSS freely and quantified intragastric administration of 10%DSS in mice.Compared with normal control groups,the model groups have shown that daily weight gain and length of colon decreased and the Disease Activity Index(DAI)increased.The mice in quantified intragastric administration group had higher frequency of occurrence of symptoms and had similar time of emergence of symptoms,lower mortality rate and less DSS consumption than those in free drinking group.It’s providing that quantified intragastrie administration of 10%DSS could induce a stably acute ulcerative colitis which is more suitable for later study.2.To investigate the therapeutic effect of RPSLP on ulcerative colitis mice.UC model mice were given intragastric administration at the dose(25 or 50 or 100 mg · kg(BW)-1,once daily,RPSLP)after 8 days DSS induced.The results showed that RPLSP blocked colon shortening,obviously reduced histological damage of colon and improved macroscopic scores,and significantly decreased contents of colonic tissue levels of TNF-α,IL-1β,IL-6 resulting from colitis of UC mice.Real time PCR(RT-PCR)results showed that RPSLP administration could down-regulate the expression of TNF-α,IL-1β,IL-2,IL-4,IL-6,iNOS mRNA,and up-regulate the expression of ZO-1,Occluding,claudin-1 mRNA in colon of mice with colitis.Above all,it proved that RPSLP was effective on treating UC mice.3.To investigate the interventional effect of RPSLP on ulcerative colitis mice.KM mice were induced by DSS,at the same time RPSLP was given at the dose(12.5 or 25 or 50 mg·kg(BW)-1,once daily)by intragastric administration.With the administration of RPSLP,the colon injury was alleviated,the ultramicro pathology damage of colonic mucosa was restored,and the degrees of inflammation were mitigated.RPSLP also blocked colon shortening,significantly decreased contents of TNF-α,IL-1β,IL-6 in colonic tissue of DSS-induced mice.Real time PCR(RT-PCR)results showed that RPSLP administration could down-regulate the expression of TNF-α,IL-1β,IL-2,IL-4,IL-6,iNOS mRNA,and up-regulate the expression of IL-10,ZO-1,Occluding,claudin-1 mRNA in colon of DSS-induced mice.Above all,it proved that RPSLP was effective on intervening DSS-induced mice. |
