Basis of topic selectionIn recent years,malignant tumor takes the first place in human disease,and lung cancer ranks the first place in this field.Up to now,the treatment of lung cancer mainly relies on drug chemotherapy,but most of the chemotherapy drugs will cause certain harm to the body,and the repeated drug resistance has become the fatal shortcoming of chemotherapy drugs in the treatment of lung cancer.Xiben pill from the "Dongyuan test prescription",the main treatment of Xiben disease,is now lung cancer.According to our previous research,we found that the prescription composition of xiben pill is in line with the relevant dialectics of lung cancer in traditional Chinese medicine,and some relevant experiments show that xiben pill can play an anti-tumor role by promoting cell apoptosis.ObjectiveTo study the antitumor effect and mechanism of xiben pill on mice with Lewis lung cancer,and to provide some basis and reference for further development of xiben pill.Methods(1)To study the main components,core targets and key pathways of xiben pill in the treatment of lung cancer by Network pharmacology,which provides a direction for further research.(2)To establish the Lewis lung cancer mouse model,and divide them randomly into different groups.After 14 consecutive days of medication,stripping the tumors of mice and determining the tumor inhibition rate of each group.And to evaluate the pharmacodynamic results of xiben pill by the above results.(3)To observed the tumor and organ cell morphology of mice;To detect the expression of Bcl-2 and caspase 3 by IHC;To detect the expression of VEGF by RT-PCR.(4)First of all,we detect the effect of Xiben pill on A549 cell viability.Secondly,we make a determination of the protein expressions of p-Akt and VEGF in A549 cells by Western Blot and RT-PCR.In the end,we take an experiment on the effect of xiben pill on HUVEC migration,invasion and lumen analogues.Results(1)This paper screened out the protein targets and pathways associated with lung cancer by network pharmacology.and the main protein targets included Akt1,VEGFA,caspase3,Bcl-2,etc.the main pathways include cancer pathway,PI3K-Akt pathway and so on.(2)Animal experiments showed that Xiben pill could inhibit tumor growth in Lewis lung cancer mice in a dose-dependent manner.(3)The cell morphology of various organs in different groups have no significant difference in the results of HE staining.Compare with the model group,and the tumor cells were much smaller and flat in the experimental group.IHC showed that Xiben pill could affect the expression of Bcl-2 and Bcl-2.RT-PCR showed that Xiben pill could inhibit the expression of VEGF.(4)This paper studied the PI3K/ Akt /VEGF pathway by cell experiments.And the results showed that Xiben pill could decrease the viability of A549 cells,inhibit the expression of p-Akt and VEGF.These results suggest that Xiben pill can exert anti-tumor effect by regulating PI3K/ Akt /VEGF signaling pathway,and Xiben pill can make the inhibition of angiogenesis by inhibiting the migration and invasion of HUVEC cells and the formation of luminal analogue.Conclusion(1)Xiben pill can indeed play an anti-lung cancer role by regulating these targets and pathways screened by network pharmacology.This shows that network pharmacology can be used in the study of the mechanism of action of Chinese herbal formula,and it has a certain guiding significance in the experiment.(2)Xiben pill can inhibit tumor growth in mice with Lewis lung cancer and thus exert anti-lung cancer effect.(3)Xiben pill can regulate PI3K/ Akt /VEGF signaling pathway.Inhibiting HUVEC angiogenesis.Thus play an anti-lung cancer role.
