As a traditional Chinese medicine prescription,Zuojin Pill is widely used in clinical treatment of gastrointestinal-related diseases.In view of its good curative effect,experts and scholars from various countries are committed to studying the broader pharmacological effects of Zuojin Pill.Animal experiments have shown that Zuojin Pill has a sustained antihypertensive effect,little toxic and side effects.However,its antihypertensive effect has not been verified in cell experiments.Therefore,in order to scientifically molecular mechanism of Zuojin Pill in treating hypertension,this research uses the current novel network pharmacology combined with cell experiments.The results of this article are described as below:1.In this research,network pharmacology method was conducted for the first time to preliminary explore the hypotensive mechanism of Zuo Jin Wan.Based on the TCMSP database,the component information of Zuojin Pill was firstly obtained,and28 candidate components were collected.The 80 potential targets associated with the candidate components were further obtained.Then,a candidate compound-predicted target network model was established through the Cytoscape software platform.Subsequently,using hypertension as a key word to search for targets that were closely related to hypertension in the database.Then a disease-target network system was built based on the Cytoscape software platform.Through mutual mapping,a network of medicine-active ingredients-key targets-disease was successfully constructed.There were 18 key targets in the network map.Moreover,the types of key targets and the importance of proteins in the interaction network were further analyzed.The resulut found that ADRA1B protein showed strong interaction with other proteins.Finally,the DAVID database was selected to obtain the biological processes and pathways involved in key targets.Network analysis revealed that Zuojin Pill’s anti-hypertensive effect may involve c GMP-PKG signaling pathway,Ca2+signaling pathway and other pathways.2.This study uses cell experiments to confirm the validity and reliability of the results of network pharmacological analysis at the cellular level.Whereafter,the effects of induction time and concentration on cell viability were investigated.The experimental results demonstrated that the 600μmol/L induction for 2 h was suitable for the establishment of EA.hy926 cell oxidative damage model.3.In this article,the extract of Zuo Jin Wan was prepared by reflux extraction.Then we explored the effects of different concentrations of Zuojin Pill solution on cell viability.Analysising the figure,the dose of 150μg/m L,100μg/m L and 75μg/m L concentration did not substantially damage the cells,so the concentration of 150μg/m L and below was used as the non-toxic dose of Zuojin Pill.Subsequently,the result of drug repair experiments on cell damage indicated that administration within 100μg/m L~150μg/m L can repair the oxidative damage of cells.So the efficacy of Zuojin Pill in the aspect of anti-hypertension has been initially confirmed from the angle of cells.
