Objective: In this study,through bilateral ovariectomy in female C57BL/6J mice and administration of Finn regimen,a HRT mice model was established to simulate human menstrual cycle.By observing the changes of mice weight,shape and weight of uterus(ovary)and the weight of subcutaneous tumor of endometrial carcinoma in HRT mice model,the effect of HRT on the proliferation of subcutaneously tumor of endometrial carcinoma was comprehended.Through the summary of Meta analysis,further compare the results of HRT model from a clinical point of view.Verify the effect and discuss the related mechanism preliminarily.Methods: 1.Study on estrous cycle and screening of visible components in vaginal secretions of C57BL/6J mice which can be used as indicators of surgical effect.For 14 consecutive days,at the same time every day,the visible components in the vaginal secretions of 14 sexually mature and non-pregnant C57BL/6J mice aged 7 to 8 weeks were examined by wet mount and classified counting.Statistics on the changes of different visible components of vaginal secretions in the corresponding time.Then they were randomly divided into two groups: control group(PC)and ovariectomized group(POVX).Bilateral ovariectomized mice in POVX group and sham operation in PC group were performed.Pap staining and classified counting of visible components in mice vaginal secretions were performed 8 days before and after operation.2.Construction of HRT model and its application in the treatment of endometrial carcinoma: 21 sexually mature and non-pregnant C57BL/6J mice aged 7 to 8 weeks were randomly divided into three groups: control group(C),bilateral ovariectomized HRT group(HRT)and bilateral ovariectomized group without HRT(OVX).The number of keratinocytes in the vaginal secretion of mice was observed within 1 cycle after operation to confirm whether the operation was successful.After confirming the success of the operation,HRT group was given hormone treatment according to Finn regimen,while C group and OVX group were given control treatment.3.Meta analysis: Through electronic database search,Chinese and English articles with NOS scores of more than 6 stars were selected according to the inclusion and exclusion criteria to collect the basic data related to the recurrence rate of postoperative application of HRT in endometrial cancer and Meta analysis,and verify the sensitivity and publication bias of the results.Results: 1.In order to reveal the estrous cycle of C57BL/6J mice in the feeding environment of this study,and the feasibility of using different visible components of vaginal secretions as detection indexes of vaginal secretions,the visible components of vaginal secretions of mice were examined by wet mount for 14 days.The results showed that the fluctuation cycle of seedless keratinized cells was 5.65 ±1.5 days,that of nucleated epithelial cells was 3.11 ±1.18,and that of leukocytes was 4.54 ±1.28.There was no obvious fluctuation cycle and quantitative change of erythrocytes,and the fluctuation cycle of seedless keratinized cells disappeared after bilateral ovariectomy.2.(1)The visible components of vaginal secretions in group C,HRT and OVX were observed continuously for 8 days.The results showed that the fluctuation cycle of the number of seedless keratinocytes still existed in group C,and disappeared in group HRT and OVX,indicating that the operation was successful.(2)There was significant difference in body weight of tumor-bearing mice without treatment(p < 0.05).The effects of different treatment factors on the body weight of tumor-bearing mice changed with time.The change range of body weight in HRT group and OVX group was the most obvious,and there was a downward trend in the later stage,while that in group C was not obvious.(3)There was no significant difference in the tumor weight of HEC-1-B endometrial carcinoma among the three groups(p > 0.05),but the tumor weight in HRT group was significantly less than that in the other two groups(0.05<p<0.1).(4)The results of killing and dissecting the mice showed that the appearance of the uterus in group C was full,the outline of the uterus was clear,and the color was normal or light red;the appearance of the uterus in the HRT group was full,the outline was clear,the size was similar to that of the control group,and the color was light red;in the OVX group,the appearance was wrinkled,atrophied to linear,and the color was pale and dull.There was significant difference in uterine weight among the three groups(p < 0.05).3.In this paper,we searched the electronic database and screened according to the inclusion and exclusion criteria,and finally included 10 Chinese and English literatures with more than 6 stars.Through the Meta analysis of these 10 articles,it was found that the use of HRT in patients with endometrial carcinoma did not increase the recurrence rate,and it was possible to reduce the postoperative recurrence rate.Conclusions: 1.In the visible components of vaginal secretions of C57BL/6J mice,nucleated keratinized cells,nucleated epithelial cells and white blood cells can reflect the changes of estrous cycle,and seedless keratinized cells can be used as a reference index of estrous cycle 3 before and after ovariectomy.using keratinized cells as a reference index of C57BL/6J,the estrous cycle in the current feeding environment is 4-7 days,with an average of5.65 ±1.5 days.2.In female C57BL/6J mice,the weight of postmenopausal mice changed rapidly after castration,and the body weight of mice decreased significantly after tumor bearing,while the change of body weight of mice in HRT group was more stable than that of uncastrated mice without hormone replacement.3.After hormone replacement with Finn regimen,it has a certain inhibitory effect on the proliferation of endometrial cancer cells.4.HRT can effectively avoid uterine atrophy and improve the quality of life of postoperative patients.5.Hormone replacement therapy may reduce the postoperative recurrence rate in patients with endometrial carcinoma.6.Through Meta analysis,postoperative HRT treatment of endometrial cancer patients,will not increase its recurrence rate,and there is a possibility to reduce its postoperative recurrence rate.
