Objective: As we all known,potential of natural antioxidants such as resveratrol in prevention and treatment of malignancies.At present,the effect and mechanism of resveratrol against bladder cancer cells have been confirmed,but its clinical application is limited due to low bioavailability.Oxyresveratrol,as a derivative of resveratrol,which biological activity is more stable than resveratrol,and it has attracted more and more attention in medical research.However,Oxyresveratrol mechanism of inhibiting bladder cancer cells is still unclear.Methods:(1)Oncomine and GEPIA databases were used to analyze the expression of EGR2 gene in muscular invasive bladder cancer and non-muscular invasive bladder cancer,and to analyze the expression of EGR2 gene in different stages and grades of bladder cancer and the influence of EGR2 gene on the survival rate of patients with bladder cancer.(2)The status of bladder cancer T24 cells treated with different concentrations of resveratrol was observed by inverted microscope.(3)CCK8 experiment was conducted to investigate the effects of different concentrations of resveratrol on the proliferation of bladder cancer T24 cells,so as to select the best concentration of resveratrol.(4)The expression of EGR2 m RNA and EGR2 protein in bladder cancer T24 cells were investigated by QRT-PCR and Western Blot assay.(5)Western Blot assay was used to investigate the effects of resveratrol on apoptosis genes such as caspase-3 in bladder cancer T24 cells.(6)The effect of resveratrol oxidation on the migration ability of bladder cancer cells was investigated by cell scratch test and migration test.(7)Invasion experiment was conducted to investigate the effect of resveratrol oxide on the invasion ability of bladder cancer T24 cells.Results:(1)Oncomine and GEPIA database analysis showed that EGR2 was expressed in all types of bladder cancer;The expression of EGR2 was positively correlated with the tumor stage and grade of patients with bladder cancer,and EGR2 was negatively correlated with the survival rate of patients with bladder cancer,suggesting that EGR2 gene plays a certain role in the occurrence and development of bladder cancer.(2)Oxidation of resveratrol inhibited the growth of T24 cells in a concentration-dependent manner.(3)Resveratrol inhibited the proliferation of T24 cells in a dose-dependent manner,and the inhibitory concentration(IC50)value of resveratrol was 530.8 μmol/L.(4)Oxidized resveratrol inhibited the expression of EGR2 in bladder cancer T24 cells.(5)Oxidized resveratrol promoted the apoptosis of T24 cells.(6)Oxidation of resveratrol inhibited the migration of T24 cells.(7)Oxidation of resveratrol inhibited the invasion of bladder cancer T24 cells.Conclusions:(1)The expression of EGR2 was positively correlated with tumor stage and tumor grade in patients with bladder cancer,and EGR2 was negatively correlated with survival rate in patients with bladder cancer.Resveratrol oxidation could inhibit the expression of EGR2 gene.(2)Oxidized resveratrol inhibited the proliferation,migration and invasion of bladder cancer T24 cells,and promoted the apoptosis of bladder cancer T24 cells.
