Background:Depression is one kind of serious psychosomatic disease. The behavior is to be depressed enduring state such as body discomfort and sleep-disorder. It affects normal life and social activity. Along with the rapid rhythm of social competition, the incidence of the disease is increasing. According to the report "disease burden"expressed by the WHO in 2001, the disability that depression brings about ranks the 1st. The disease burden that depression brings about ranks the 4th.It will rise to second to be next only to heart disease. It is called the second killer of the health in the next century. Undoubtedly, the research of the depression is an important problem which is faced with the medical researchers. The acupuncture has already got the affirmative curative effect on the depression, which is welcomed by the patients.Objective:In order to take advantage of the acupuncture---our traditional medical methods, this thesis is focused on observing the effects on the depressing rats by blood-letting on the hand twelve wells point .It make clear that this method has the validity on depression. Further more, it discusses this method the effects on the levels of IL-1βand IL-6 in plasma of depressing rats, in order to the mechanism of the acupuncture.Methods:32 wistar rats are randomly allocated to four groups: control group, model group, fluoxetine group and blood-letting group, each group has 8 rats. Depression model is established by using chronic unpredictable mild stimulation combined with lonely feed, continuously for21days, exceptthe control group. The fluoxetine groups are applied irritating stomach by 2ml brine which fluoxetine 2.8 mg/kg dissolves, everyday, for 21days. Blood-letting group are punctured on twelve wells point, every two days, 11 times in total. Bleeding amounts are 1 drop per point. On the 22nd day, we use ELISA method to determine the levels of IL-1βand IL-6 in plasma of depressing rats.Results:1. Compared with the model group, the level and perpendicularity score of control group have notable difference (P <0.01); the blood-letting group and the fluoxetine group have less notable difference (P <0.05 ~ 0.01); The blood-letting group and the fluoxetine group have no notable difference (P> 0.05).2.Compared with the model group, the levels of IL-1βand IL-6 in plasma of control group have notable difference (P <0.01); the blood-letting group and the fluoxetine group have notable difference (P <0.01); The blood-letting group, the model group, and the fluoxetine group have no notable difference (P> 0.05).Conclusion:1. The experiment combining the chronic unpredictable mild stimulation with lonely feed sets up the depressing animal model successfully, which provides experimental platform to the further research..2. The fact that blood-letting on the hand twelve wells points can affect the rats'behavior, has improved its curative effect on the depression.3. The immune cell change may be one of the mechanisms of depression.4. The effects of the blood-letting maybe have something with the level of IL-1β, IL-6 in the plasma.5. This thesis suggests firstly that blood-letting on hand twelve wells points has significant guidance in clinical treatment. |
