Study On The Anti-tumor Effect Of Xiaoaiping Injection Combined With Paclitaxel On Ovarian Cancer Transplanted Tumor In Zebrafish

Ovarian cancer is the malignant tumor with the highest mortality among female reproductive organ tumors,which is a great threat to the life and health of many women.The onset of ovarian cancer is hidden and is not easy to be found.Once found,it is basically at an advanced stage.Conventional treatment of ovarian cancer includes surgery,radiotherapy,chemotherapy,targeted therapy and immunotherapy.Chemotherapy plays a leading role in the treatment of ovarian cancer.However,due to the multidrug resistance of tumor cells and the side effects of radiotherapy and chemotherapy,it is difficult for advanced cancer and elderly patients to adhere to it.Therefore,the search for new tumor therapeutic drugs has become an urgent problem to be solved.Traditional Chinese medicine has become the focus of antineoplastic drug research because of its multiple targets and low price.Xiaoaiping injection is a sterilized aqueous solution made from the dried rattan stem of Marsdenia tenacissimae by water extraction and alcohol precipitation.It can inhibit the proliferation and migration of tumor cells by inhibiting tumor cell proliferation,promoting tumor cell apoptosis,inhibiting tumor neovascularization,regulating immunity and so on.Clinically,it is often used alone or in combination with chemotherapy and radiotherapy for the treatment of many kinds of tumors.As a model of drug screening,zebrafish has been widely used in the research and development of antineoplastic drugs because of its similar physiological and biochemical characteristics to mammals,low cost,high efficiency and rapidity.Objective:To evaluate the anti-tumor effects of Xiaoaiping injection combined with paclitaxel on ovarian cancer transplanted tumor in zebrafish,so as to provide new idea for the prevention and treatment of ovarian cancer.Methods:CM-Dil fluorescence labeled human ovarian cancer A2780 cells(or A2780/taxol cells)were microinjected into the yolk sac of zebrafish to establish a zebrafish ovarian cancer transplant tumor model.The fluorescence intensity of zebrafish transplanted tumor was collected under fluorescence microscope.The growth rate of tumor inhibited by drugs was calculated.The incidence of tumor metastasis was calculated.The inhibitory effect of tumor metastasis was calculated by the cumulative distance of tail metastasis of zebrafish tumor cells to judge whether Xiaoaiping injection combined with paclitaxel have potential anti-tumor effect on ovarian cancer.Results:Both Xiaoaiping 1.25ng/piece and paclitaxel lOng/piece alone significantly inhibited the growth of A2780 transplanted tumor,while Xiaoaiping 0.625 and 1.25ng/piece,paclitaxel 0.625,2.5,5 and lOng/piece alone significantly inhibited the metastasis of A2780 cells.The combination of Xiaoaiping 0.625,1.25ng/piece and paclitaxel 5ng/piece significantly inhibited the growth of A2780 transplanted tumor,and Xiaoaiping 0.156,0.3125,0.625 and 1.25ng/piece combined with paclitaxel 5ng/piece significantly inhibited the metastasis of A2780 cells.Xiaoaiping alone and paclitaxel alone had no significant inhibitory effect on the growth of A2780/taxol transplanted tumor.The combination of Xiaoaiping and paclitaxel 7.5ng/piece had no significant effect on the metastasis of A2780/taxol cells.Conclusion:Both single and combination of the two drugs significantly inhibit the growth and metastasis of human ovarian cancer cell line(A2780 cell).The combination of the two drugs has a synergistic anti-tumor effect on human ovarian cancer sensitive cell line A2780,which shows a significant inhibitory effect on the metastasis of human ovarian cancer transplanted tumor in zebrafish.The combination of the two drugs had no significant inhibitory effect on the growth and metastasis of drug-resistant human ovarian cancer cell line(A2780/taxol cell).
