| Objective: Through the research of aconite-dried ginger for chronic ulcerative colitis mucosa microcirculation improving function and influence on related immune inflammatory factor,clarify aconite-dried ginger the mechanism of action of drugs for the treatment of chronic ulcerative colitis,for clinical use of aconite-dried ginger medicine to provide theoretical basis for the treatment of chronic ulcerative colitis,provide new way for traditional Chinese medicine treat chronic ulcerative colitis.Methods: Male C57BL/6 mice were selected and divided into 6 groups according to random number table method,with 10 mice in each group: normal group,model group,Guben yichang group,and aconite-dried ginger high-dose,medium-dose and low-dose groups.During modeling,the normal group was given drinking water,and the other groups were given 2%DSS aqueous solution for 5 days.On the 6th day,free drinking water was used for 5 days.This is 1 cycle,and this is repeated for 3 cycles,for a total of 30 d.During the establishment of the model,the DAI score increased significantly,indicating that the model of chronic ulcerative colitis was established successfully.After the model is built,The Guben Yichang group was given 1.16g/kg by gavage,the aconite-dried ginger high-dose,medium-dose and low-dose groups were given 6.06,3.03 and 1.51g/kg by gavage,respectively,and the normal group was given equal volume of distilled water by gavage.The dose volume was 0.2 m L/10 g,and the mice were given intragastric administration for 10 days,once a day.The body weight,stool characteristics and blood in stool were recorded every day,and the DAI score was performed.After the last administration,the blood perfusion of intestinal mucosa,the velocity of blood cells and the concentration of mobile blood cells were observed by laser Doppler flowmeter.Colon length of each group was measured by ruler.The degree of colonic mucosal inflammation and injury was observed by naked eyes and the CMDI score was performed.He staining was used to observe the pathological sections of colons in each group.Serum levels of IL-1β,IL-6,IL-10,PAF and TF were detected by enzyme-linked immunosorbent assay.The expression of NF-κB and CD31 in colon tissue of mice in each group was detected by immunohistochemical method,and the results were analyzed.Results:1.In terms of the effect of aconite-dried ginger on general symptoms of intestinal mucosa in mice with chronic ulcerative colitis,compared with model group,the DAI score of high,medium and low dose groups was significantly decreased(P <0.01);The colonic length of mice in the high-dose and medium-dose groups was significantly improved by aconite-dried ginger medicine(P < 0.01),and the colonic length of mice in the low-dose group was improved by aconite-dried ginger medicine,but the difference was not statistically significant.The CMDI score of colon mucosa of mice in high and medium dose groups was decreased by aconite-dried ginger medicine(P < 0.01),but there was no significant difference in the improvement of CMDI score of mice in low dose group.The results of HE staining showed that the colonic mucosa of mice in normal group was intact and the glands were arranged neatly.Model group mice colonic mucosa of inflammatory cell infiltration,arrange badly damaged glands,level is not clear,serious,hyperplasia of lymphoid tissue abnormal intestinal epithelial cell regeneration,aconite,dried ginger medicine of high,medium and low dose group mice colonic mucosa goblet cells increased obviously,inflammatory cells infiltration decreased significantly,glandular structures tend to be more complete,hyperplasia tissue decreased significantly.2.From the effect of aconite-dried ginger on intestinal mucosal microcirculation in chronic UC mice,compared with the model group,aconite-dried ginger medicine of high,middle dose group mice intestinal mucosal blood flow perfusion,blood move speed increased(P < 0.01 or P < 0.05),aconite-dried ginger medicine of high dose group mice intestine mucosa mobile blood concentration decreased(P < 0.01),aconite-dried ginger in the dose group mice intestine mucosa mobile blood concentration differences had no statistical significance(P > 0.05);The content of serum PAF in high-dose,medium-dose and low-dose groups was decreased in different degrees by aconite-dried ginger(P < 0.01 or P < 0.05),and the content of serum TF in high-dose,medium-dose and low-dose groups was also decreased in different degrees by aconite-dried ginger(P < 0.01).The expression of CD31 in intestinal mucosa endothelial vessels of high-dose and medium-dose groups was significantly increased(P < 0.01 or P < 0.05),but there was no significant change in low-dose group(P > 0.05).Through microscope observation,the colonic structure and intestinal villus margin defects of model group mice were observed.The brown particles with positive CD31 expression were unevenly distributed in the central axis of intestinal villus,and the number of particles was small.There were more brown granules and more granules in colonic mucosa of mice in high and medium dose groups.3.From the effect of aconite-dried ginger on the expression of inflammatory indicators in mice with chronic ulcerative colitis,compared with the model group,aconite-dried ginger of high,middle,low dose group mice serum IL-1β content has the varying degree decreased(P < 0.01),aconite-dried ginger of high,middle dose group of mice have different levels of serum IL-6 content decreased(P <0.01),aconite-dried ginger of low dose group there was no statistically significant difference of serum IL-6 levels in mice.The content of serum IL-10 in high-dose and medium-dose groups was increased by aconite-dried ginger(P < 0.01),but there was no significant difference in the content of serum IL-10 in low-dose group by aconite-dried ginger(P > 0.05).The expression of NF-κB in colon tissue of high-dose and medium-dose groups was significantly decreased(P < 0.01),but there was no significant difference in low-dose group(P > 0.05).Observation under microscope showed that there were more brown particles in the cytoplasmic nucleus of the colon tissue of mice in the model group,while the brown particles in the cytoplasmic nucleus of the colon tissue of mice in the high-dose,medium-dose and low-dose groups of aconite-dried ginger were significantly reduced.Conclusion:1.Aconite-dried ginger medicine can improve the general symptoms of chronic ulcerative colitis,and has a significant improvement effect on the colonic injury and pathological changes of chronic ulcerative colitis.2.Aconite and dried ginger medicine can improve the intestinal mucosal microcirculation disturbance in chronic ulcerative colitis.3.Aconite and dried ginger medicine can improve the intestinal mucosal microcirculation disturbance in chronic ulcerative colitis. |
