| Bergenia crassifolia(L.)Fritsch belongs to the Saxifraga family,and it is a perennial herb with a unique growth environment.In China,it is mainly distributed in Xinjiang.Bergenia crassifolia(L.)Fritsch is a characteristic medicine of the Kazakh nationality.It has the effects of relieving cough and asthma,stopping bleeding,and curing cervical erosion.Various studies have shown that it also has a variety of pharmacological activities including protecting the liver,kidney and stomach,anti-tumor,anti-bacterial,anti-inflammatory and anti-parasitic effects.In this paper,we summarized the research on the chemical constituents and biological activities of Bergenia crassifolia(L.)Fritsch and systematically investigated the chemical constituents of Bergenia crassifolia(L.)Fritsch.In addition,the immune activity of the chemical constituents is preliminary studied.After using cold-impregnation with 70% ethanol and ultrasonic assisted extraction,Organic solvents sequential polar extraction from low to high polarity,Silica gel column chromatography,gel(Sephadex LH-20,MCI)column chromatography,polyamide column chromatography,AB-8 macroporous resin column chromatography,silica gel thin layer chromatography and recrystallization methods,24 monomer compounds were separated and identified from Bergenia crassifolia(L.)Fritsch.Seven flavonoids were identified through the physical and chemical state of the compound,the thin layer,the nuclear magnetic resonance spectrum data and the literature comparison: quercetin-3-O-β-D-xyloside(9),quercetin(14),kaempferol(15),kaempferol-3-O-L-arabinofuranoside(16),(-)-epicatechin(17),quercetin-3-O-α-L arabino-furano side(19),(-)-epigallocatechin-3-O-gallate(21),5 sterol compounds: β-sitosterol(3),stigmasterol(4),betulinic acid(6),oleanolic acid(18),carotin(12),3 glycoside compounds: arbutin(10),6-O-galloyl arbutin(11),rhamnose(23),2 organic acid compounds: ursolic acid(7),gallic acid(13),two coumarin compounds: bergenin(8),11-O-gallocylbergenin(20),two fatty compounds: thirtydodecane(1),tetracosanoic acid(2),and 3 other compounds: hydroquinone(5),quincelic acid(22),mannitol(24).Among them,the compounds 2,9,16,19,21,23,24 were isolated from Bergenia plants for the first time,and the compounds 2,4,6,7,9,16,17,18,19,21,22,23,24 were isolated from Bergenia crassifolia(L.)Fritsch for the first time.Eight kinds of monomer compounds extracted from Bergenia crassifolia(L.)Fritschcan have the effect of promoting the splenic lymphocyte transformation of mice.Quercetin(14)and kaempferol(15)can promote splenic lymphocyte transformation at a lower concentration of 2 μg/m L.Hydroquinone(5),quercetin-3-O-β-D-xyloside(9),6-O-galloylarbutin(11),and epicatechin(17)can promote lymphocyte transformation at 5 μg/m L.Quercetin-3-O-ɑ-L-arabino furanoside(19)and epigallocatechin gallate(21)can promote splenic lymphocyte transformation at 10 μg/m L and 20 μg/m L in a dose-dependent manner.Among them,the promoting effect of quercetin-3-O-β-D-xyloside(20 μg/m L)on the splenic lymphocyte transformation was the most obvious. |
