Objective:Determining the expression of the related proteins caspase-1,IL-1β,NLRP3 and GSDMD-N in breast cancer tissues and adjacent tissues,and analyzing the relationship between the expression of the corresponding protein and the clinicopathological parameters of breast cancer,as well as the relationship between the effector protein of the pyrolysis pathway.To explore the relationship between the expression of caspase-1,IL-1β,NLRP3 and GSDMD-N and the prognosis of breast cancer patients,in order to provide new strategies for molecular targeted therapy of breast cancer patients.Methods:Collected 108 archived breast cancer(Invasive breast cancer,not a special type)specimens(paraffin embedded)and 40 paracarcinoma tissue specimens confirmed in our hospital from January 2014 to December 2014,and the immunohistochemical Elivision TM plus method was used to measure the production of caspase-1,IL-1β,NLRP3 and GSDMD-N in 108 breast cancer tissues and 40 benign lesion tissues adjacent to cancer.Results:(1)The expressions of effector proteins caspase-1,IL-1β,NLRP3 and GSDMD-N in the pyrolysis way in breast cancer tissues were significantly different from those in adjacent tissues(P<0.05),And the expression of caspase-1,IL-1βand NLRP3 protein in breast cancer tissues is generally lower than that of adjacent tissues,and the expression of GSDMD-N protein in breast cancer tissues is higher than that of adjacent tissues.(2)Correlation analysis of caspase-1,IL-1β,NLRP3 and GSDMD-N in 108 breast cancer tissues showed that caspase1 and IL-1β were significantly positively correlated(r=0.609,P<0.001);caspase1 and GSDMD-N is a significant positive correlation(r=0.549,P<0.001);caspase1 and NLRP3 are significantly positively correlated(r=0.566,P<0.001);IL-1β and GSDMD-N are significantly positively correlated(r= 0.410,P<0.001);IL-1β is significantly positively correlated with NLRP3(r=0.396,P<0.001);GSDMD-N is significantly positively correlated with NLRP3(r=0.334,P<0.001).(3)In 108 breast cancer tissues,the higher the expression of caspase-1,IL-1β,GSDMD-N and NLRP3 protein,the lower the histopathological grade of breast cancer tumor,the smaller the tumor size,the lower the clinical stage,and the more likely lymph node metastasis.Low,the lower the risk of death,the better the prognosis,the difference is statistically significant(P<0.05),the protein expression of caspase-1,IL-1β,NLRP3 and GSDMD-N are not significantly correlated with the patient’s age(P>0.05)).(4)The COX multi-factor model was used to carry out the research,and the outcome manifested that the low or no expression of caspase-1,IL-1β,NLRP3 and GSDMD-N protein,clinical stage,pathological grade,tumor magnitude and lymphatic metastasis are independent elements affecting the survival of breast cancer patients danger factors.(5)The Kaplan-Meier survival analysis was used to analyze the survival of 108 breast cancer patients.The results showed that the overall survival time of patients in the high expression group of caspase-1,IL-1β,NLRP3 and GSDMD-N protein was significantly higher than that in the low expression group.There are differences Statistically significant(P<0.05).Conclusions:(1)The down-regulation of caspase-1,IL-1β and NLRP3 expression may be related to the occurrence of breast cancer,the low expression of GSDMD-N protein in the adjacent cancer indicates that under normal circumstances,the adjacent tissues do not initiate or rarely initiate the pyrolysis process,but in breast cancer tissues,the pyrolysis pathway will be activated and lead to the occurrence of pyrolysis.(2)In breast cancer,caspase-1,IL-1β,NLRP3 and GSDMD-N proteins work together to initiate pyrolysis.(3)Relation analyse,COX retuen analyse and subsist analyse of caspase-1,IL-1β,NLRP3 and GSDMD-N and clinicopathological arguments of breast cancer patients showed that caspase-1,IL-1β,NLRP3 and GSDMD-N may play an important role in the invasion,metastasis and prognosis of breast cancer.
