| Objective: In this study,we will investigate the relationship between NLRP3 inflammatory bodies and non-obstructive coronary microvascular disease(NOCMVD),by comparing NLRP3 protein and mRNA expression in peripheral blood mononuclear cells(PBMCs)and IL-18 and IL-1β expression in plasma between healthy people and patients with NOCMVD.Methods:50 NOCMVD patients were selected from the Cardiovascular Center of Cardiovascular and Cerebrovascular Hospital,Yan ’an University Affiliated Hospital from January 2019 to June 2020 as the experimental group,and 50 healthy subjects in our hospital were selected as the control group.the basic clinical data and blood-related indicators of all participants were collected.10 ml of blood was collected from the pulmonary vein of all participants before eating in the morning,and the plasma and peripheral blood mononuclear cells were separated and stored in-80℃ refrigerator.NLRP3 protein and mRNA expression in PBMCs were detected by Western blot and RTPCR,respectively,and IL-18 and IL-1β expression was determined by ELISA.By performing statistical analysis,the relationship between NLRP3 inflammatory bodies and non-obstructive coronary microvascular disease was investigated.Results:1.When the basic clinical data of the patients in the two groups were compared,only the history of hypertension was statistically significant(P < 0.05).2.Comparison of blood related indicators between the two groups showed no statistical difference(P > 0.05).3.The expression of plasma IL-18 in the experimental group was(124.82±28.04)ng/L,which was significantly increased compared with(85.10 ±28.39ng/L)in the control group,and the expression of IL-1 β in the experimental group was(25.38±8.53)ng/L,was significantly increased compared with(15.95 ±8.41ng/L)in the control group(P < 0.05).4.NLRP3 protein expression in PBMCs was significantly increased in the experimental group(0.99 ± 0.40)compared with the control group(0.41 ± 0.21)(P < 0.05),and NLRP3 mRNA expression in PBMCs was significantly increased in the experimental group(1.94 ± 0.76)compared with the control group(0.93 ± 0.50)(P < 0.05).5.Logistic regression analysis showed that the expression of IL-18,IL-1β,NLRP3 protein,and NLRP3 mRNA were independent risk factors for NOCMVD(P < 0.05).6.The predictive value of IL-18,IL-1β,NLRP3 protein,and NLRP3 mRNA expression for the occurrence of NOCMVD was analyzed using ROC curves,and the results showed that the areas under the curve were 0.834,0.784,0.907,and 0.858 respectively,the sensitivities were 0.80,0.70,0.78,0.76,and the specificities were 0.72,0.74,0.88 and 0.84,respectively.It is suggested that the expression of IL-18,IL-1β,NLRP3 protein and NLRP3 mRNA had good value in predicting the occurrence of NOCMVD.7.It was found that the occurrence of NOCMVD was positively correlated with plasma IL-18 and IL-1β expression,NLRP3 protein,and NLRP3 mRNA expression in PBMCs,by using Spearman correlation analysis,with correlation coefficients of 0.492,0.579,0.705,and 0.621,respectively(P < 0.05).8.Pearson correlation analysis1)Plasma IL-18 expression was positively correlated with NLRP3 protein and NLRP3 mRNA expression in patients of the experimental group,with correlation coefficients of 0.581 and 0.681,respectively,(P < 0.05)。2)There was a positive correlation between plasma IL-1β expression and NLRP3 protein and NLRP3 mRNA expression in PBMCs in the experimental group,respectively,with correlation coefficients of 0.717 and 0.682,respectively,(P < 0.05).Conclusion:1.The occurrence of NOCMVD is associated with increased expression of NLRP3 protein,NLRP3 mRNA in PBMCs,and IL-18 and IL-1β in plasma.2.NLRP3 protein,NLRP3 mRNA in PBMCs,and IL-18 and IL-1β in plasma all have good value in predicting the occurrence of NOCMVD. |
