| Objective:In this study,SD rats were used as the research object,and the obstructive jaundice liver injury model was created by ligation of the common bile duct,and modified Dachaihu decoction was used as an intervention to explore the correlation of the IRE1α-ASK1-JNK pathway in the endoplasmic reticulum stress pathway.It provides a theoretical basis for the clinical application of modified Dachaihu decoction to alleviate liver function impairment and perioperative period in patients with obstructive jaundice.Methods:Ninety SD male rats were divided into sham operation + normal saline group(group S,n = 30);OJ model + normal saline group(group O,n = 30);OJ model + modified Dachaihu decoction group(group M,n = 30).Groups O and M were surgically ligated into the common bile duct,entered the abdominal cavity through the mid-abdomen along the xiphoid process,free the bile duct and double-ligated the common bile duct with a sterile surgical suture;group S only entered the abdomen from the same site,and the free common bile duct was not ligated.From the first day after surgery,group S and group O were given intragastrically once a day with normal saline(1ml /100g);group M were given once a day with modified dachaihu decoction(MMDB)(1ml / 100g).Each group was sampled at 3,7,and 10 days,including heart blood and liver tissue,each time Ten rats were randomly selected in the group.HE staining method was used to observe the pathological condition of rat liver tissue;biochemical analysis instruments were used to detect the changes of serum aspartate aminotransferase(AST),alanine aminotransferase(ALT)and serum total bilirubin(TBIL)in each group;RT-PCR and Western Blot detection of JNK and Bcl-2 m RNA and protein expression expression in rat liver tissue.Results:1.HE staining of the pathological changes of the liver tissues of rats: the liver cells in group O and M were indistinct,the liver cell cord arrangement structure was disordered,the hepatic sinusoid dilatation,cholestasis,cell degeneration and necrosis.There was no obvious pathological change in group S.2.Changes in serological indicators: compared with group S,the serum AST,ALT and TBIL contents in group O and group M were all increased,which was significantly higher than that in group and the difference was statistically significant(P < 0.05).Compared with group O,the serum levels of AST,ALT and TBIL in group M were lower than that in group O at each time point,and the difference was statistically significant(P < 0.05).3.RT-PCR and Western Blot detection of JNK and Bcl-2 m RNA and protein expression changes in rat liver tissue: β-actin as the internal reference protein,compared with group S,JNK m RNA and protein expression level in group O and M were higher(P<0.05);in group O were higher than those in group M at each time point,the difference was statistically significant(P <0.05);in groups O and M Bcl-2 m RNA and protein expression levels were lower than those in group S(P <0.05);in group O were lower than those in group M at each time point,and the differences were statistically significant(P<0.05).Conclusions:The rat model of obstructive jaundice was successfully established by ligating the common bile duct of rats using traditional surgical methods.When modified Dachaihu decoction is applied to the endoplasmic reticulum stress hepatocyte apoptosis caused by obstructive jaundice rat model,its mechanism may be to down-regulate the expression of JNK protein and promote the expression of Bcl-2 to inhibit hepatocytes the process of apoptosis,so that the liver function of rats is improved.For the clinical treatment of patients with obstructive jaundice,in addition to the appropriate surgical method is the guarantee,modified Dachaihu decoction can be used by TCM differentiation of syndromes to reduce the damage of liver function in patients with obstructive jaundice.Benefit from this and provide new treatment ideas. |
