| ObjectiveTo observe the effect of acupuncture and Moxibustion"Feishu","Dazhui"and"Fengmen"on the expression of substance P and vasoactive intestinal peptide protein in the lung and large intestine of chronic asthma model rats,to explore the mechanism of“Shao’s Method of Wu Zhen”in the treatment of chronic asthma model rats and its regulatory effect on intestinal histopathology and related regulatory substances,and to explore the law of"lung disease and intestine"transmission,so as to provide a new way for“Shao’s Method of Wu Zhen”in the treatment of pulmonary enteropathy and enteropathy.To observe the effect of acupuncture and Moxibustion"Feishu","Dazhui"and"Fengmen"on the expression of substance P and vasoactive intestinal peptide protein in the lung and large intestine of chronic asthma model rats.Secondly,the mechanism of“Shao’s Method of Wu Zhen”in the treatment of chronic asthma model rats,its regulatory effect on intestinal histopathology and related regulatory substances were comprehensively investigated.Finally,a novel therapy of“Shao’s Method of Wu Zhen”was provided to treat pulmonary enteropathy and enteropathy by exploring the law of"lung disease and intestine transmission".Method40 healthy male SPF SD rats were randomly divided into blank group,model group,acupuncture group and moxibustion group.On the first and eighth day of the experiment,rats in the model group,acupuncture group and moxibustion group were intraperitoneally injected with 10%compound egg protein antigen solution(1ml)for sensitization.From the 15thday of the experiment,1%egg protein solution was inhaled by atomization to excite for 20min(once every other day for 6 weeks)to establish the chronic asthma rat model.The rats in the blank group were injected intraperitoneally with normal saline instead of ovalbumin.Acupuncture group was treated with acupuncture,moxibustion group was treated with moxibustion,and rats in both groups were treated with"Feishu"(double)"Dazhui","Fengmen"(double)"Zusanli"(double),which was stimulated after treatment.The blank group received grasping stimulation without any other treatment.After the last treatment,the lung and colon tissues were extracted and fixed with 4%paraformaldehyde.Paraffin embedding was used to prepare pathological sections,hematoxylin eosin(HE)staining was used to observe the pathological morphology of lung and intestine,and immunohistochemical staining was used to observe the expression of SP and VIP protein in lung and intestine.Results1 Analysis of behavioral signs of rats in each groupThe rats in the blank group had good mental state,stable breath and normal stool.In the model group,the rats were coughing,sneezing,shortness of breath,scratching their ears and scratching their cheeks,fidgeting,slow response,dry stool and less.Compared with the model group,acupuncture group and moxibustion group showed better mental state,more stable breathing and sensitive response,and then their stool tended to be normal.2 Morphological observation of lung tissue in each groupIn the blank group,the lung tissue structure was intact and clear,the alveolar space was normal,the bronchial lumen was regular,and there was no obvious inflammatory cell infiltration in the interstitial lung.In the model group,the local alveolar structure was disordered,a small number of exfoliated epithelial cells can be seen in the lung interstitium,a large number of inflammatory cells were infiltrated in and around the bronchial lumen,the wall of the tube was hyperplasia,and the lumen was narrow.Acupuncture group and moxibustion group:the alveolar wall was basically complete,the smooth muscle of bronchus was slightly thickened,and a small number of inflammatory cells were infiltrated in and around the bronchus lumen.3 Observation of intestinal histomorphology of rats in each groupIn the blank group,the structure of intestinal mucosa epithelium was intact,the large intestine gland was clear,and there was no inflammatory cell infiltration.In the model group,the intestinal mucosa epithelium was not complete,the arrangement of glands was not regular,a small amount of mucosa epithelium fell off,and inflammatory cells wereinfiltrated in submucosa,muscular layer and serosa layer.In the acupuncture group and moxibustion group,the intestinal mucosa epithelium was relatively complete,and a smallnumber of inflammatory cells infiltrated.4 Comparison of SP protein expression in lung tissue of rats in each groupThe expression of SP protein in the lung tissue of the model group was significantly higher than that of the blank group(P<0.01);the expression of SP protein in the lung tissue of the acupuncture group and the moxibustion group was significantly lower than that of the model group(P<0.01);there was no the statistically significant difference between the acupuncture group and the moxibustion group.(P>0.05).5 Comparison of VIP protein expression in lung tissue of rats in each groupThe expression of VIP protein in the lung tissue of the model group was significantly lower than that of the blank group(P<0.01);the expression of VIP protein in the lung tissue of the acupuncture group and the moxibustion group was significantly higher than that of the model group(P<0.01);the difference between the acupuncture group and the moxibustion group was not statistically significant(P>0.05).6 Comparison of SP protein expression in intestinal tissue of rats in each groupThe expression of SP protein in the intestinal tissue of the model group was significantly lower than that of the blank group(P<0.01),and the expression of SP protein in the intestinal tissue of the acupuncture group and the moxibustion group was significantly higher than that of the model group(P<0.01);the difference between the acupuncture group and the moxibustion group was not statistically significant(P>0.05).7 Comparison of VIP protein expression in intestinal tissue of rats in each groupThe expression of VIP protein in the model group was significantly higher than that in the blank group(P<0.01).The expression of VIP protein in the acupuncture group and moxibustion group was significantly lower than that in the model group(P<0.01).There was no significant difference between the acupuncture group and moxibustion group(P>0.05).Conclusion1“Shao’s Method of Wu Zhen”can effectively improve the symptoms and behavioral signs of chronic asthma model rats.2“Shao’s Method of Wu Zhen”can effectively improve the pathological morphology of lung tissue and intestinal tissue in rats with chronic asthma.3“Shao’s Method of Wu Zhen”downregulate the expression of SP protein and upregulate the expression of VIP in lung tissue of rats with chronic asthma.Acupuncture and moxibustion regulating the imbalance of SP and VIP may be one of the ways to treat asthma.4“Shao’s Method of Wu Zhen”can upregulate the expression of SP protein and downregulate the expression of VIP in intestinal tissue of rats with chronic asthma.5 In the state of asthma,lung disease can affect the intestine,which changes the pathological morphology and related regulatory substances of the lung and intestine.Acupuncture and moxibustion treatment of asthma may also improve intestinal disease,and achieve the effect of lung and intestine treatment. |
