ObjectiveObserve effects of Shao’s “Wu Zhen ”Method on Eotaxin and CCR3 of lung tissue of asthmatic rats in each group.Investigate Shao’s “Wu Zhen ”Method on asthma to provide more scientific basis for acupuncture clinic.MethodForty rats were divided into four groups randomly:blank,model,drug and acupuncture group.Establish asthmatic rats with ovalbumin and aluminum hydroxide.Acupuncture asthmatic rats in Fei Shu(BLI3,double),Da Zhui(DU14,double),Feng Men(BL12,double)in acupuncture group daily.Inject through intraperitoneal 0.05% dexamethasone sodium phosphate in drug group daily.No treatment after stimulation in model group.Inject through intraperitoneal and inhalate with saline instead of ovalbumin in blank group.After the last treatment taking out the lower part of the right lung tissue.Then Prepare biopsy.Count Eos percentage ang observe morphological changes of lung tissue through HE staining.Immunohistochemistry is used to detect Eotaxin and CCR3 in lung tissue of rats.Results 1 The behavior of rats ach groupModel group appeared shortness of breath,lethargy compared with control group;Rats of blank group had no obvious abnormalities;Shortness of breath and other symptoms of drug and acupuncture group reduced compared with model group.2 Lung tissue morphology each groupAirway of rats of blank group structural integrity,no significant damage,no inflammatory.Airway epithelial of model group damaged loss,partially visible mucus plug,submucosal edema,smooth muscle proliferate;surrounding interstitial lung airways lot infiltration of inflammatory cells.Airway of drug and acupuncture group basic integrity,less inflammatory in lung tissue,structure was normal.3 Eos count of each groupEos percentage of model group was more than blank group,difference was obvious(P<0.05);acupuncture and drug group were less than model group,difference was obvious,but more than blank group;acupuncture group comparing with drug group,difference was obvious.4 Comparison of Eotaxin each groupModel group showed a larger number of Eotaxin compared with blank group,difference was obvious(P<0.05);Eotaxin of acupuncture and drug group decreased compared with model group.Difference was obvious and acupuncture group was less than drug group.Difference was obvious;blank group was with little expression.5 Comparison of CCR3 each groupModel group was seen a lot of CCR3.Compared with blank group,difference was obvious(P<0.05);acupuncture and drug group,CCR3 reduced compared with model group.Difference was obvious.And acupuncture group was less than drug group;blank group was with little expression.6 Correlation AnalysisCorrelation analysis showed that Eos percentage of lung tissue was highly positive correlated with Eotaxin and CCR3 expression(r = 0.826,0.805).Conclusion 1 Shao’s “Wu Zhen ”Method could ameliorate symptoms of asthmatic rats.2 Shao’s “Wu Zhen ”Method could lessen lung tissue morphology of asthmatic rats.3 Shao’s “Wu Zhen ”Method could lessen Eos infiltration in lung tissue of asthmatic rats.4 Shao’s “Wu Zhen ”Method could lessen Eotaxin and CCR3 in lung tissue of asthmatic rats.5 Chemotaxis of Eos to lung tissue maybe related to Eotaxin and CCR3.Shao’s “Wu Zhen ”Method may inhibit Eotaxin and CCR3 of Eos to chemotaxis lung tissue,thereby reducing Eos inflammatory in lung tissue to achieve the purpose of treatment of asthma.
