| Aconitum is a processing product of aconitum root of the family buttercup.First recorded in the”Shen Nong Ben Cao Jing”,has the strong property of great heat and great toxicity.Modern scholars believe that the effect of aconite on strengthening the heart is the main pharmacological effect of the clinical efficacy of traditional Chinese medicine（returning to Yang to save the life,supplementing fire to help Yang）.Aconitum is a kind of processed product in clinical it is toxic.Jiangxi Jianchang gang to the processing of aconite unique,Yin-fu tablets,Yang-fu tablets is especially characteristic."Jianchang Bang Paozhi Quanshu"records Yin-fu tablets,Yang-fu tablets because of its different processing methods and differences in drug use,Yin-fu tablets is suitable for female kidney yang deficiency,Yang-fu tablets is suitable for male kidney yang deficiency,there is no record of gender differences in the treatment of chronic heart failure.On the basis of the previous research,in this paper,the effects of Yin-fu tablets and Yang-fu tablets on heart-strengthening in rats with chronic heart failure were studied systematically,to compare the effect of Yin-fu tablets and Yang-fu tablets on chronic heart failure,in order to provide theoretical basis for clinical application of Yin-fu tablets and Yang-fu tablets.This study mainly studies the effect of Yin-fu tablets and Yang-fu tablets on the strengthening heart and the mechanism of strengthening heart in rats with chronic heart failure,whether there are gender differences.A rat model of chronic heart failure was established by intraperitoneal injection of doxorubicin gradient,detection of hemodynamic parameters of rats in each group by biological function signal acquisition system,HE stained sections were used to observe the pathology of myocardial tissue in rats.To explore the effect of Yin-fu tablets and Yang-fu tablets on adriamycin-induced chronic heart failure rats.Turns out,the hemodynamic parameters of rats with chronic heart failure Left ventricular systolic pressure（LVSP）,left ventricular end-diastolic pressure（LVEDP）,maximum increase and decrease rate(±LVd P?dtmax-1)of left ventricular internal pressure were significantly decreased in rats with chronic heart failure.Yin-fu tablets showed the most significant increase in d P?dtmax-1in female rats and Yang-fu tablets showed the most significant increase in d P?dtmax-1in male rats.Yin-fu tablets and Yang-fu tablets can effectively improve the pathological conditions of vacuole and widening rupture of tissue space in myocardial tissue of heart failure rats.Then,the mechanism of action of Yin-fu tablets and Yang-fu tablets on heart failure rats was developed from four aspects,modern drugs for the treatment of heart failure are mainly to alleviate the inflammation of heart failure,regulate the RAAS system of heart failure patients,improve myocardial energy metabolism,and regulate the expression of epinephrine receptors.So this experiment through the study of these four pharmacological indicators to explore the mechanism of its action.1.Study on inflammatory response of Yin-fu tablets and Yang-fu tablets in rats with heart failure,serum levels of interleukin-6（IL-6）,brain natriuretic peptide（BNP）,interleukin-10（IL-10）,tumor necrosis factor-α（TNF-α）were determined by enzyme-linked immunosorbent assay.Results the IL-6、BNP、TNF-αcontents of Yin-fu tablets and Yang-fu tablets decreased,IL-10 content increased.Indicating that Yin-fu tablets and Yang-fu tablets can reduce the level of serum inflammatory factors,improve the level of anti-inflammatory factors,and alleviate the inflammatory response in vivo.The IL-6、BNP of Yang-fu tablets decreased significantly in male rats,Yin-fu tablets decreased most significantly TNF-αfemale rats.2.Study on regulation of RAAS system in rats with heart failure by Yin-fu and Yang-fu tablets,serum renin（Renin）,angiotensin II（AngⅡ）and aldosterone（ALD）levels were determined by enzyme-linked immunosorbent assay,RT-PCR expression levels of angiotensin type I receptor（AT1R）m RNA in myocardial tissue of rats were measured,the protein expression levels of AT1R in myocardial tissue of each group were detected by Western blotting method.Results the serum Renin、Ang-Ⅱ、ALD,myocardial tissue AT1Rm RNA expression and AT1R protein expression were significantly increased in rats with chronic heart failure.Serum Ang-Ⅱ、ALD levels decreased significantly in rats of Yin-fu tablets and Yang-fu tablets group,Renin downward trend,and the levels of AT1Rm RNA expression and protein decreased significantly.Yang-fu tablets showed the most significant decrease in Renin、ALD content in male rats,and Yin-fu tablets showed the most significant decrease in AT1Rm RNA expression level and protein expression level in rats.3.Effects of Yin-fu tablets and Yang-fu tablets on myocardial energy metabolism in rats with heart failure,the content of myocardial tissue of cells ATP and the activity of Na-K-ATP、Mg2-ATP、Ca2-ATP enzyme were measured ATP chronic heart failure rats by colorimetry.As a result,the ATP content and enzyme activity decreased significantly in the model rats,ATP content increased and enzyme activity increased in the group of Yin-fu tablets and Yang-fu tablets.The activity of Mg2-ATP Yang-fu tablets was significantly higher than that of Yin-fu tablets in male rats,and that Na-K-ATP Yin-fu tablets was significantly higher than that of Yang-fu tablets in female rats.4.Effects of Yin-fu tablets and Yang-fu tablets on adrenergic receptor expression in heart failure rats,the expression level of epinephrineβ1,β2 receptor in myocardial tissue m RNA rats was detected by RT-PCR method.Experimental results showed that the expression ofβ1 receptor increased significantly in the model rats,β2 receptor expression decreased significantly,Yin-fu tablets and Yang-fu tablets can reduceβ1receptor expression,enhanceβ2 receptor expression.Yin-fu tablets to decreased expression ofβ1 receptor in female rats was the most significant,the results showed that the protective effect of the Yin-fu tablets on myocardium of female rats was the most significant.In summary,effect of Yin-fu tablets and Yang-fu tablets on chronic heart failure rats,its mechanism is not single.Are mainly to alleviate the inflammatory response,inhibit the excessive activation of the RAAS system,improve myocardial energy metabolism,and regulate the expression of adrenaline receptors in four aspects.In terms of heart strong,the improved ventricular diastolic function Yin-fu tablets was significantlyhigher than that of Yang-fu tablets in female rats,the ventricular systolic function Yang-fu tablets was significantly higher than that of Yin-fu tablets in male rats.In reducing inflammatory response,decreased IL-6,BNP of Yang-fu tablets on male rats significantly.And in inhibiting RAAS overactivation of the body,Yin-fu tablets to decreased AT1Rm RNA expression level and protein expression was most significant in different sex rats.Regulation of adrenergic receptor expression,Yin-fu tablets decreased expression ofβ1 receptor in female rats was the most significant.There are some gender differences in the therapeutic effect of Yin-fu tablets and Yang-fu tablets on chronic heart failure rats. |
