Research Background:Polycystic ovary syndrome(PCOS)is a common endocrine disorder in women of childbearing age.It is mainly manifested as reproductive disorders and metabolic abnormalities.Modern medical treatment of PCOS is mostly symptomatic,and hormone drugs,PCOS are mostly used.It is a chronic disease that requires long-term medication.Western medicine has many side effects and the condition is easy to repeat after withdrawal.In recent years,there have been more and more studies on the treatment of this disease by Chinese medicine.After many years of clinical research,the tutor has applied the homemade Zishen recipe to the clinic,and the clinical observation has obvious curative effects,but the molecular mechanism of its possible role remains to be explored.Follicular developmental disorders are the main manifestation of PCOS.Granulosa cells(GC)are an important part of follicular development.Mature follicles require continuous differentiation and proliferation.However,PCOS patients have fewer GCs than normal,and GC layers shrink and thin This may be the main cause of ovulation disorders in patients with PCOS.Therefore,on the basis of clinically proven efficacy,to explore the specific mechanism of action between Zishen recipe and PCOS granule cells from the perspective of basic experiments will be of great significance to the occurrence and development of disease.Objective:1.Through clinical research,this topic observes the effect of Zishen Decoction on BMI,blood hormones and lipid metabolism related indexes in patients with PCOS with kidney yin deficiency,and provides clinical basis for the later treatment of PCOS by traditional Chinese medicine.2.Based on previous clinical research,carry out cell experimental research to explore the possible mechanism of Zishen recipe on PCOS.method:1.Select 35 patients with PCOS who met the inclusion criteria in the outpatient clinic of Jiangxi University of Traditional Chinese Medicine and conducted self-control study.Oral Chinese medicine compound treatment for 3 months,observe the changes of BMI,blood hormones and lipid metabolism indexes before and after treatment.2.Select 45 female SD rats weighing 200-260 g,and divide the rats into two groups,a traditional Chinese medicine group and a blank group,with20 rats in each group,and the remaining 5 rats will be spared;In the stomach for 14 days,the blank group was administrated with physiological saline at a ratio of 2ml / 100 g body weight;the Chinese medicine group was administrated with Chinese hormone index improved significantly.The levels of LH,LH / FSH,and T in blood decreased significantly before treatment compared with after treatment,and there was a significant statistical difference(P<0.01).;The level of FSH in blood was higher before treatment than after treatment,although the difference was not statistically significant(P>0.05);the level of E2 in blood was significantly higher before treatment than after treatment,and there was a significant statistical difference(P <0.01).2.After 3 months of treatment with Zishen Decoction,the body mass index BMI level of patients decreased compared with that before treatment,and there was a significant statistical difference(P <0.01).3.After 3 months of treatment with Zishen Decoction,the patient’s lipid metabolism index improved significantly.The levels of TG and LDL before treatment were significantly lower than those after treatment,with a statistically significant difference(P <0.01);HDL levels before treatment.Compared with that after treatment,the increase was obvious,and there was a significant statistical difference(P <0.01).4.C / EBPβ was expressed in both groups of GC,and the expression of C / EBPβ m RNA in GC of the Chinese medicine group was significantly higher than that of the blank group,with a significant statistical difference(P <0.01).5.Compared with the blank group,the number of cells in the Chinese medicine group was significantly increased after 48 hours,and the OD values were higher from 1d to 7d.conclusion:1.Zishen Decoction can effectively improve the related indexes of sex hormones,BMI,and lipid metabolism in patients with PCOS with kidney yin deficiency,thereby having a therapeutic effect on PCOS;2.Zishen Decoction may promote the proliferation of PCOS granule cells by up-regulating the expression of C / EBPβm RNA,so as to effectively treat PCOS disease.This may be one of the possible mechanisms of Zishen Decoction for PCOS disease.
