The Effects Of IFN-γ On Ovarian Granular Cells In Polycystic Ovary Syndrome

ObjectivePolycystic ovary syndrome(PCOS)is an endocrine disordered disease in women of reproductive age,which accompaning abnormal follicular development,obesity,compensatory hyperinsulinemia,hyperandrogenemia.It has been reported that chronic inflammation is closely related to PCOS.In this study,the levels of IFN-γin PCOS patients were detected and investigated its roles in the pathogenesis and development of polycystic ovary syndrome(PCOS).Methods1.Peripheral blood of 31 patients with PCOS and 22 healthy volunteers were collected,the levels of T、E2、LH、PRL、FSH、P in these samples were detected. In addition,peripheral blood mononuclear cells(PBMCs)were harvested from these samples,and the proportions of IFN-γ~+Th1 and IL-4~+Th2 cells in PBMCs were analyzed using flow cytometer.2.The levels of IL-4、IL-6 and IFN-γin plasma of PCOS patients were detected bythe FCM CBA method.3.Human granulosa-like tumor cell line KGN cultured with IFN-γin vitro,theKGN cells were divided into two groups,control group(routine culture)andIFN-γtreaded group(treaded with 250 ng/mL IFN-γ).The effects of IFN-γonthe cell cycle,proliferation and apoptosis of KGN cells were analyzed using flowcytometer.4.Culture supernatant of control group and IFN-γtreatment group were collectedand the levels of T、E2、LH、FSH、P、AMH in supernatant were detected.Results1.Compared with the healthy control group,The levels of T、E2 and LH in PCOS patients were significantly higher than that in the control group(P<0.01),and the level of PRL was increased in PCOS patients(P<0.05).The proportion of Th1 cells in PBMCs of PCOS patients was significantly up-regulated(P<0.01).2.The levels of IL-6 and IFN-γin plasma of PCOS patients were higher than the control group(P<0.05).3.After being co-cultured IFN-γwith KGN cells in vitro,the results showed that the proliferation of CFSE labeled KGN cells in the IFN-γtreaded group was significantly inhibited comparing with the control group.Both the percentage of KGN cells in G0-G1 phase and the proportion of apoptotic KGN cells increased in the IFN-γtreaded group(P<0.01).4.The levels of T、E2、LH、FSH、P、AMH between the control group and the IFN-γ treatment group were no difference in culture supernatant.Conclusion1.The levels of androgen and the proportion of IFN-γ~+Th1 cells in PBMCs of peripheral blood were increased in PCOS patients.2.The levels of IL-6 and IFN-γwere increased in PCOS patients.3.IFN-γinhibited the proliferation,but promoted the apoptosis of KGN cells.
