Objective: To analyze the composition and the diversity of microbiome of adenoid in children using 16 SrDNA gene sequencing and bacterial culture,and to evaluate the resistance of pathogenic bacteria of adenoid in children by drug sensitivity test.Methods: Samples of adenoid secretions were obtained from 25 adenoid hypertrophy children and 6 healthy children.16 SrDNA gene-sequencing technology was used to analyze the bacterial community composition,alpha diversity,beta diversity and other biological information of these samples.The bacterial community composition and diversity between the two groups and each children were analyzed and compared.At the same time,these samples were cultured for aerobic bacteria,anaerobic bacteria and fungus.The susceptibility of the cultured pathogenic bacteria was tested.Samples of adenoid secretions and adenoid tissue were collected from 108 children who undergoing adenoidectomy.These samples were cultivated for aerobic bacteria,anaerobic bacteria and fungi,and the susceptibility of the cultured pathogenic bacteria was tested too.The differences of bacteria communities between the surface and tissue of adenoid were analyzed,as well as the resistance of pathogenic bacteria in adenoids.Results: The predominant bacterial communities of adenoid hypertrophy children were Streptococcus,Fusobacterium,Prevotella,Haemophilus,Moraxella,etc.There was an observable difference in the relative abundance of bacterial communities between adenoid hypertrophy children.The group of adenoid hypertrophy children had higher bacterial community diversity and abundance compared to healthy children group(chao1 index and simpson index,P<0.05,respectively).The predominant pathogenic bacteria colonies cultivated were Streptococcus pneumoniae,Haemophilus influenzae,Staphylococcus aureus,etc.These pathogenic bacteria colonies always resist to several antibiotics simultaneously.Conclusion: Our results indicate children’s adenoid bacterial communities contain multiple bacterial colonies.These bacterial colonies are keeping balance in healthy children,while the balance state was destroyed in children with adenoid hypertrophy.And the diversity of adenoid bacterial colonies are high among individuals of adenoid hypertrophy children.The pathogenic bacteria colonies are highly resistant to antibiotics.
