Objective: The purpose of this experiment was to observe the general condition,disease activity index(DAI)and interleukin-17(IL-17),interleukin-23(IL-23),peroxisome proliferator activated receptor-γ(PPAR-γ),and nuclear factor-B(NF-κB)of ulcerative colitis(UC)rats treated by huazhong jiedu decoction,so as to explorethe possible mechanism of huazhuojiedu decoction in the treatment of ulcerative colitis.Methods: Forty 8-week-old male Wistar rats were randomly divided into a blank group(NG)of 10 and a modeling group of 30.The modeling group was used to induce the UC rat model by sodium dextran sulfate(DSS)method.After 10 days of continuous modeling,27 UC model rats successfully prepared were divided into model group(MG),western medicine group(XG)and traditional Chinese medicine group(HG)according to the random number table method.The blank group and model group were gavaged with normal saline,and the other treatment groups were gavaged with corresponding drugs for 14 consecutive days.To observe the general condition of hair,movement,eating,defecation and the changes of colonic mucosal histopathology.The levels of IL-17 and IL-23 in serum of rats were detected by enzyme-linkedimmunosorbent assay(ELISA),and the expressions of PPAR-γ,NF-κ B in colonic tissues were detected by immunohistochemical assay.Results:1 General condition and histopathological observation of ratsNG rats were generally in good condition,with no abnormal colon tissue under the microscope.The activity,feeding and water intake of MG rats decreased,and their body weight decreased.The rats became thin or saw pus and blood in mucus,and the glands were significantly damaged under the microscope,with a large number of inflammatory cells infiltrating.The general condition of the rats in each treatment group was improved,and the colonic mucosal tissue structure was relatively neat under XG microscope,with a small amount of inflammatory cell infiltration.The mucosal glands of the HG colon are more regular,and scattered inflammatory cells can be seen in each layer.2 DAI scoreCompared with NG,DAI scores of the rats in each group increased(P<0.05).Compared to MG,DAI scores of XG and HG rats were decreased,with statistically significant differences(P < 0.05).There was no significant difference between XG and HG(P>0.05).3 Serum IL-17 test results of rats in each groupCompared with NG,serum IL-17 content of MG rats was significantly increased,with statistically significant difference(P<0.05).Compared with MG,serum IL-17 content of rats in each treatment group was significantly decreased,with statistically significant difference(P < 0.05).There was nosignificant difference between XG and HG(P>0.05).4 Serum IL-23 test results of rats in each groupCompared with NG,serum IL-23 content of rats in other groups increased,with statisticallysignificant difference(P<0.05).Compared with MG,serum IL-23 content of rats in each treatment group was decreased,with statistically significant difference(P<0.05).Compared with HG and XG,serum IL-23 content was significantly decreased,with statistically significant difference(P <0.05).5 Expression of PPAR-γin colonic tissues of rats in each groupCompared with NG,the expression of PPAR-γ in the colon tissues of MG rats was significantlydecreased,with statistically significant results(P<0.05).Compared with MG,the expression of PPAR-γ in colonic mucosa of rats in the treatment group was increased,with statisticallysignificant results(P<0.05).There was no significant difference between XG and HG(P<0.05).6 Expressionof NF-κB in colonic tissues of rats in each groupThe statistical results showed that compared with NG,the content of NF-κB in XG colon tissues increased,with no significant difference(P>0.05),while the expression of NF-κB in other groups increased,with statistically significant difference(P<0.05).Compared with MG,NF-κB expression in colonic mucosa of XG and HG rats decreased,with statistically significant results(P<0.05).Compared with HG,XG decreased the expression of NF-κB in colon tissues,and the difference was statistically significant(P <0.05).Conclusion:1 Huazhuojiedu decoction can improve the general condition of UC rats and alleviate the pathological injury of UC rats.2 Huazhuojiedu decoction may play a role in the treatment of UC by upregulating the expression of PPAR-γ,inhibiting the activation of NF-κB,IL-17,IL-23 and other inflammatory factors,thereby reducing and controlling the inflammatory reaction of the colon mucosa,preventing tissue inflammation and mediating self-tolerance,so as to regulate the immune status of the body.
