| [Background and Objective]Postpartum Depression(PPD)is the most common type of depression in women,and it is a major hazard to the health of babies and mothers.At present,western medicines commonly used in clinical treatment of PPD have certain limitations and difficulties.Guhan Yangshengjing is a national key protected traditional Chinese medicine variety.In recent years,some studies have found that the components of Guhan Yangshengjing also play a wide role in depression,but no studies have directly reported that Guhan Yangshengjing can treat PPD.This experiment used network pharmacology to predict the effective components and possible targets of Guhan Yangshengjing in the treatment of PPD,and further explored the effects of Guhan Yangshengjing on HIF-1α signal pathway,inflammatory response and synaptic plasticity in the hippocampus of PPD rats in the PPD animal model,so as to explain the mechanism of Guhan Yangshengjing in the treatment of PPD and provide evidence for Guhan Yangshengjing in the treatment of PPD.[Methods]1.Network pharmacology analysis: tcmsp database and genecards database were used to screen the target genes related to Guhan Yangshengjing and PPD,and the key targets were selected.Then the core components of Guhan Yangshengjing were analyzed.Finally,the mechanism of Guhan Yangshengjing in the treatment of PPD was obtained through GO function analysis and KEGG pathway analysis;2.PPD modeling and grouping:The female SD was subjected to an open field experiment(OFT)to screen qualified rats,and they were randomly divided into: control group(CG group),model group(MG group),Guhan Yangshengjing low-dose group(G-L group),middle-dose group(G-M group)and high-dose group(G-H group).The model group and Guhan Yangshengjing group were treated with bilateral ovariectomy,continuous hormone injection and withdrawal of hormone after simulated pregnancy to prepare PPD model rats.After modeling,OFT,TST and FST were used to evaluate the model.3.Use WB to detect related targets:After 2 weeks of gavage treatment with Guhan Health Essence,the hippocampus tissue of the rat was decapitated and the hippocampus tissue of the PPD model rat was studied by Western blotting to detect the expression of candidate target genes obtained through network pharmacology screening.[Results]1.Through the analysis of network pharmacology,it is found that Guhan Yangshengjing is a combination of multiple active components,among which flavonoids may be the core components in the treatment of PPD,such as kaempferol,beta sitosterol,isorhamnetin,etc;2.The active components of Guhan Yangshengjing can act on multiple targets,mainly through IL6 and HIF-1α、Jun,STAT3 and other targets are the core targets of Guhan Yangshengjing in the treatment of PPD;3.Guhan Yangshengjing can treat PPD through multiple pathways,the key pathways including HIF-1 signaling pathway,AGE-RAGE signaling pathway,AMPK signaling pathway and FOXO signaling pathway,etc;4.Guhan Yangshengjing can inhibit the expression of HIF-1α in the hippocampus of PPD rats;5.Guhan Yangshengjing can reduce the expression of IL-6inflammatory factors in the hippocampus of PPD rats;6.Guhan Yangshengjing can promote the expression of synaptic function protein PSD-95 in PPD rats.[Conclusion]Through the analysis of network pharmacology,it shows that the mechanism of Guhan Yangshengjing in the treatment of PPD has the characteristics of "multi-component,multi-channel and multi-target";Guhan Yangshengjing can inhibit the production of HIF-1α in the hippocampus of PPD model rats,inhibit hippocampal inflammation,thereby improving hippocampal synaptic plasticity. |
