Objective: Based on network pharmacology,to explore the mechanism of Huanglian jiedu decoction in the treatment of hypertension,evaluate its clinical efficacy in the treatment of hypertension(grade 1),and verify the key target of network pharmacology.to provide new ideas for the treatment of essential hypertension with traditional Chinese medicineMethods: 1.Network pharmacology research: a total of 429 ingredients of Huanglian jiedu decoction were searched from TCMSP database.After the screening of ADME parameters,a total of 86 active components were found to be in line with oral bioavailability(OB)and drug(DL)after weight removal.1662 drug targets were retrieved,and 144 drug targets were obtained after standardization and weight removal by UNIPROT.Related targets were obtained by searching the disease target database as follows: TTD 81,OMIM 200,Dis Ge NET 445,CTD,111431 and Gene Cards 8435.According to the correlation criteria,17777 and 1325 targets were screened respectively.A total of 1845 hypertension-related targets were obtained by UNIPROT standardization and EXCEL software combined with weight removal.110 biological processes related to protein binding,cytokine activity and antioxidation were obtained by GO analysis,and112 pathways were found between fluid shear stress and atherosclerosis,AGEs-RAGE signal pathway,calcium signal pathway,c GMP-PKG signal pathway,IL-17 signal pathway and so on.2.Clinical study: a total of 60 patients with hyperactivity of liver fire in accordance with essential hypertension(grade 1)were randomly divided into treatment group(n = 30)and control group(n = 30).The treatment group was treated with Huanglian jiedu decoction and the control group was treated with valsartan capsule.The antihypertensive effect,the improvement of TCM syndromes,safety,blood glucose,blood lipids,inflammatory indexes and the changes of key targets in serum were evaluated.Results: 1.Network pharmacology research: a total of 429 ingredients of Huanglian jiedu decoction were searched from TCMSP database.After the screening of ADME parameters,a total of 86 active components were found to be in line with oral bioavailability(OB)and drug(DL)after weight removal.1662 drug targets were retrieved,and 144 drug targets were obtained after standardization and weight removal by UNIPROT.A total of 20592 related targets were obtained by searching the disease target database.According to the correlation criteria,a total of 1845 hypertension related targets were obtained by UNIPROT standardization and EXCEL software combined with weight removal.GO analysis showed 110 biological processes related to protein binding and antioxidation,and 112 pathways related to fluid shear stress and atherosclerosis.2.Clinical study: Huanglian jiedu decoction can reduce blood pressure,and its antihypertensive effect is similar to that of valsartan capsule.It can improve TCM syndrome,improve blood glucose,TG,TC,LDL-C and HDL-C,and reduce inflammatory indexes such as HS-CRP,VEGFA,PTGS2,NOS3 and IL-6.It has good security.Conclusions: 1.The mechanism of Huanglian jiedu decoction in the treatment of hypertension may be through fluid shear stress,atherosclerosis,AGEs-RAGE signal pathway,regulation of key targets such as IL-6,regulation of inflammatory factors,vasomotor level,metabolism and endocrine level to achieve the purpose of regulating hypotension mechanism.2.The clinical efficacy and safety of Huanglian jiedu decoction in the treatment of essential hypertension(grade 1)with liver fire hyperactivity syndrome should be affirmed.
