Objective:In this paper,the components of chloroform fraction of Dalbergia benthamii Prain.were separated and identified,and then quality control and analysis,which be used to make a draft quality standard for Dalbergia benthamii Prain.Methods:1 Chemical InvestigationThe chloroform fractions of Dalbergia benthamii Prain.were separated and purified by preparative reversed-phase high performance liquid chromatography.The monomer compounds were identified and separated by relevant method.2 Quality Control of Dalbergia benthamii Prain.According to the method prescribed in the Chinese Pharmacopoeia of 2015 edition,the moisture,ash and acid insoluble ash and the extraction results in the medicinal materials of Dalbergia benthamii Prain.from different areas were determined.The TLC was used to identify the chemical constituents of Dalbergia benthamii Prain.and different habitats.The different habitats fingerprints of Dalbergia benthamii Prain.was established by HPLC.The common pattern was established by the similarity evaluation system of chromatographic fingerprints.The corresponding results were analyzed by cluster analysis machine principal component analysis combined with SPSS(21 edition).The contents of Dalbergia benthamii Prain.were determined by HPLC.On this basis,to develop a method of quantitative analysis of multi-components by single marker(QAMS)for simultaneously determining six constituents in Dalbergia benthamii Prain.Results:1 Chemical InvestigationFour compounds were isolated from chloroform fractions of Dalbergia benthamii Prain.2 Quality Control of Dalbergia benthamii Prain.The TLC identification could be used for qualitative identification of Dalbergia benthamii Prain.The HPLC fingerprint of Dalbergia benthamii Prain.has 14 common peaks,six components were identified by HPLC-DAD,with the similarities over 0.9.Twelve habitat can be divided into two categories by SPSS cluster analysis.There are three principal components in principal component analysis.The results of principal component rotation matrix show that4′-hydroxy-5,7-dimethoxy-6-(3-methyl-2-butenyl)-isoflavone is the most contribution in principal component analysis.Based on the single factor study,the optimum extraction conditions of Dalbergia benthamii Prain.were determined.The results showed that the precision of the instrument was good,and the solution of the sample was basically stable within 12 hours.Linear study showed that the six components was good in the range.The correlation coefficients were higher than 0.999.Conclusions:In this paper,the quality standard of Dalbergia benthamii Prain.was established by the TLC and the HPLC.The results of moisture content,ash content,acid insoluble ash content and leachate content were reasonable.Among them,the TLC method is simple,feasible,accurate and credible,and can be used for qualitative identification.The fingerprint and content determination method by HPLC have good reproducibility,high accuracy and sensitivity,and can be used for qualitative and quantitative determination of Dalbergia benthamii Prain.This standard can systematically and normatively control the quality of medicinal materials of Dalbergia benthamii Prain.by muti-indicators,and provide certain scientific basis for the development and utilization of medicinal materials resources in the future.
