Objective: Through the network pharmacology methods of biological networks,bitter and treat breast cancer research sophora active ingredients and action mechanism for the treatment of breast cancer.Methods: The pharmacokinetic parameters set as standard,in the TCMSP database screening of the chemical composition of sophora,at the same time in predicting TCMSP database screen out sophora chemical potential targets;choose Gene Cards database for breast cancer-related targets of retrieval,delete duplicate data.Made by Venny 2.1.0 software composition of sophora targets and disease targets between Wayne figure,and get the intersection genes,both for potential targets for sophora role in breast cancer.Select Cytoscape software to "Chinese medicine-chemical-targets-disease" network building,after analyzing the network topological structure,determine the active components of bitter and treatment of breast cancer and targets;using the STRING database construction of breast cancer related PPI network;application of Auto Dock Vina and molecular docking software on conformation rate and draw heat maps,verify the reliability of the key chemical composition combined with core targets;Using the R Programming Language to do the GO functional enrichment analysis and the KEGG biological pathway enrichment analysis about targets for the sophora treatment of breast cancer,illustrate the mechanism of action of bitter and treatment of breast cancer.Results: Sophora mainly through five key chemical composition,as luteolin,quercetin,phaseolin,formononetin and 8-isopentenylkaempferol,acting on the four core targets JUN,IL-6,AKT-1,MAPK1 play a role for the treatment of breast cancer.GO enrichment results showed that target genes of Sophora for breast cancer were mainly enriched in 20 biological processes,mainly focus on: steroid hormone response,lipopolysaccharide reaction,bacteriogenic molecular reactions,nutrient level response,metal ion reaction,antibiotic reaction,acid chemical reactions,ketones reaction,oxidative stress and so on.KEGG enrichment results screening had 20 pathway,it was speculated sophora could play a role in the treatment of breast cancer by influencing the fluid shear stress and atherosclosis-related signaling pathways,hepatitis B related signaling pathways,proteoglycan related signaling pathways in cancer,age-rage signaling pathways associated with diabetes complications,and herpes virus infection signaling pathways associated with Kaposi’s sarcoma.Conclusion: This research methods to explore the application of network pharmacology of sophora the function mechanism of the treatment of breast cancer after it is concluded that the main active components in sophora luteolin,quercetin etc;the main targets were Jun,JUN,IL-6,AKT-1,MAPK1;participate in process such as steroids,lipopolysaccharide;the main signaling pathways and fluid shear stress is associated with atherosclerosis,hepatitis way,second,it was associated with proteoglycan associated signaling pathways in cancer,AGE-RAGE associated with diabetes complications,and herpes virus infection associated with Kaposi’s sarcoma.For the mechanism of action of bitter and treat breast cancer research and drug development provides a certain reference. |
