The Effect Of High Expression Of LncRNA RNF157-AS1 On Cancer Cell Function In Ovarian Cancer | | Posted on:2022-07-19 | Degree:Master | Type:Thesis | | Country:China | Candidate:C Zhu | Full Text:PDF | | GTID:2504306335951489 | Subject:Obstetrics and gynecology | | Abstract/Summary: | PDF Full Text Request | | Objective: LncRNA RNF157-AS1 was identified from the cancer-related lncRNA database as experimental subjects.In vitro experiments were conducted to investigate the effect of the altered expression of lncRNA RNF157-AS1 on the biological behavior of ovarian cancer cells.Bioinformatics analysis was used to predict the possible presence of relevant targets of lncRNA RNF157-AS1.Methods: Through GEPIA,Cancer RNA-seq,lncRNAdiseases cancer-related lncRNA lncRNA of ovarian Cancer screening in network database,ovarian tissues from patients with high-grade serous ovarian cancer and ovarian cyst in Yijishan Hospital of Wannan Medical College from 2019 to 2020 were collected as experimental clinical samples,in the sample through the q RT-fluorescent quantitative PCR detection results of lncRNA RNF157-expression analysis of AS1,The differential expression of lncRNA RNF157-AS1 in ovarian cancer tissues and A2780 cells was confirmed.After the expression of lncRNA RNF157-AS1 in A2780 cells was knocked down by si RNA transfection,the functional changes of ovarian cancer cells were observed by scratch test,proliferation test and invasion test.The relevant target that may exist in lncRNA RNF157-AS1 is predicted by GEPAIA and lnc NADISeases.Results:1.The search results of Cancer RNA-seq,GEPIA and lnc Rn NADISEAS websites showed that the expression of lncRNA RNF157-AS1 was increased in ovarian Cancer tissues,P <0.01.2.The increased expression of lncRNA RNF157-AS1 in serous ovarian cancer tissue and ovarian cancer cell A2780 was statistically significant(P <0.01).3.The results of scratch test,Transwell test and proliferation test showed that knockdown of lncRNA RNF157-AS1 could inhibit the proliferation and invasion ability of ovarian cancer cells.4.Bioinformatics analysis showed that RNF157 and FOXJ1 were closely related to lncRNA RNF157-AS1.Conclusion:1.The expression of lncRNA RNF157-AS1 was significantly increased in serous ovarian cancer tissue and ovarian cancer cell A2780.2.Downregulation of the expression of lncRNA RNF157-AS1 can inhibit the migration,invasion and proliferation of ovarian cancer cells,which indicates that lncRNA RNF157-AS1 may play a pro-cancer role in ovarian cancer and may become a potential therapeutic target and prognostic marker of ovarian cancer.3.LncRNA RNF157-AS1 may interact with RNF157 and FOXJ1 to play a cancer-promoting role. | | Keywords/Search Tags: | Ovarian cancer, LncRNA RNF157-AS1, Cell proliferation, Cell invasion | PDF Full Text Request | Related items |
