| Objective: To clarify the expression and location of Wnt7 B andβ-catenin in human brain tissues and glioma tissues.To explore the effects of different concentrations of Corilagin on Wnt7 B and β-catenin in U251 glioma stem cells(U251 GSCs).Methods:1.From 1 September 2019 to 31 August 2020,21 glioma tissues from part of patients who underwent surgical treatment in the Department of Neurosurgery,Affiliated Hospital of Jining Medical University were selected as the experimental group.At the same time,12 brain tissues of patients who underwent internal decompression surgery due to traumatic brain injury were selected as the control group.The expression and location of Wnt7 B and β-catenin in glioma tissue and brain tissue was analyzed by immunofluorescence staining,immunohistochemical staining and Western Blot.2.Human U251 glioma cells were routinely cultured,and U251 GSCs were isolated,cultured and identified.The expression of Wnt7 B and β-catenin in U251 glioma cells and U251 GSCs were analyzed by Western Blot.U251 GSCs were treated with different concentrations(0μg/ml,100μg/ml,200μg/ml,400μg/ml)of Corilagin for 48 hours.The morphological changes of U251 GSCs were observed under an inverted phase-contrast microscope,the proliferation of glioma U251 GSCs were detected by CCK-8 assay,and the expression of Wnt7 B and β-catenin in U251 GSCs were detected by Western Blot.Results:1.Wnt7 B and β-catenin were both expressed in glioma tissues and brain tissues.The protein expression levels of Wnt7 B and β-catenin in glioma tissues were both significantly higher than that in brain tissues(P<0.01).Wnt7 B was mainly expressed in the cytoplasm,and β-catenin was mainly expressed in the cytoplasm and nucleus in glioma tissues.2.Wnt7 B and β-catenin were both expressed in U251 glioma cells and U251 GSCs.The protein expression level of Wnt7 B in U251 GSCs were significantly higher than that in U251 glioma cells(P<0.01).The protein expression level of β-catenin in U251 GSCs were higher than that in U251 glioma cells(P<0.05).3.After U251 GSCs were treated with different concentrations of Corilagin for 48 hours,the stem cell spheres were destroyed and cell fragments were seen in the culture medium.The survival rate of U251 GSCs gradually decreased,with the increasing concentration of Corilagin(P<0.01).4.After U251 GSCs were treated with different concentrations of Corilagin for 48 hours,the protein expression levels of Wnt7 B andβ-catenin in U251 GSCs were gradually decreased with the increasing concentration of Corilagin(P<0.01).Conclusion:1.Wnt7 B and β-catenin were expressed in human brain tissues,glioma tissues,U251 cells and U251 GSCs.Wnt7 B was mainly expressed in the cytoplasm,and β-catenin was mainly expressed in the cytoplasm and nucleus in glioma tissues.2.The protein expression levels of Wnt7 B and β-catenin in glioma tissues were both higher than that in brain tissues.The protein expression levels of Wnt7 B and β-catenin were both overexpressed in U251 GSCs compared with U251 glioma cells.3.The proliferation of U251 GSCs could be significantly inhibited by Corilagin.The protein expression levels of Wnt7 B and β-catenin in U251 GSCs were gradually decreased with the increasing concentration of Corilagin.We can inferred that Corilagin inhibited U251 GSCs through inhibiting the expression of Wnt7 B and β-catenin.But the specific mechanism needs to be further elucidated. |
