ObjectiveBipolar Disorder(BD)is a chronic mental Disorder in which the emotional state fluctuates periodically between negative(depression)and positive(mania).Many genetic and environmental factors influence the risk of bipolar disorder,but the cause remains unknown.Glycogen Synthase Kinase-3(GSK-3)is found in eukaryotic cells,is a serine/threonine Kinase and has two subtypes,GSK-3α and GSK-3β.GSK-3α is expressed only in the hippocampus,cerebral cortex,striatum,and cerebellum of the nervous system,while GSK-3β is expressed in almost all regions of the brain.The two subtypes are highly homologous,but have different functions.Previous studies on GSK-3 and bipolar disorder have mostly focused on the GSK-3β subtype.The activity of GSK-3β was affected by phosphorylation at two different sites,in which phosphorylation of Ser at 9 inactivated the enzyme and phosphorylation of tyrosine at 216 increased the activity of the enzyme.GSK-3β is a major regulator of neuronal development and morphogenesis.It interacts with Wnt/β-catenin,PI3K/Akt and other signaling molecules to regulate various cellular functions.Studies have shown that GSK-3β may play an important role in the pathogenesis of bipolar disorder.Used in the treatment of bipolar disorder,emotional stability agent lithium salt has been confirmed for the activation of GSK-3 inhibitor,GSK-3 beta is to influence the pathogenesis of bipolar disorder specificity of protein,the results still exist differences both at home and abroad,most clinical studies in patients with bipolar mania episode,very few of bipolar depression and bipolar disorder remission patients with GSK-3 beta research.In this study,peripheral blood was extracted to compare p-Ser9-GSK-3β/ total GSK-3β with normal monocytes in different clinical stages of bipolar disorder,i.e.,manic episode,depressive episode,and remission,to preliminarily explore the role of GSK-3β in the occurrence and development of bipolar disorder.Materials and methodsAccording to the group and exclusion criteria,the selection between September 2019 and September 2020 in Dalian 7 th People’s Hospital in 90 cases of patients with bipolar disorder(manic and depressive episode group,relief groups(30 cases)and 30 cases of healthy volunteers(control group)as the research object,to collect general information,assessment of manic scale(BRMS),24 Hamilton depression rating scale(HAMD),using Western Blot method(Western Blot),p-Ser9-GSK-3β/total GSK-3β in peripheral blood were detected in bipolar disorder manic group,depression group,remission group and control group,and the data were processed by SPSS 26.0 statistical software.Results1.The ratio of p-Ser9-GSK-3β to total GSK-3β in peripheral blood monocytes in bipolar mania group,bipolar depression group and bipolar remission group was higher than that in normal control group,with statistical significance(P=0.000<0.01).2.p-Ser9-GSK-3β/total GSK-3β in peripheral blood of bipolar mania and bipolar depression group was higher than that of bipolar remission group,and the difference was statistically significant(P=0.000<0.01).3.There was no significant difference in p-Ser9-GSK-3β/total GSK-3β in peripheral blood monocytes between bipolar mania group and bipolar depression group(P= 0.231>0.05).4.The ratio of p-Ser9-GSK-3β/ total GSK-3β was positively correlated with the manic scale score in bipolar mania group(R=0.37,P=0.044<0.05).There was no significant correlation between p-Ser9-GSK-3β/total GSK-3β ratio and Hamilton Depression Scale score in bipolar depression group(R=0.229,P=0.223>0.05).5.There was no significant correlation between p-Ser9-GSK-3β/total GSK-3β ratio and course of disease,age and age of onset in bipolar mania group and bipolar depression group(P>0.05).Conclusion1.GSK-3β may play an important role in the pathogenesis of bipolar disorder.2.p-Ser9-GSK-3β/total GSK-3β in peripheral blood may be a biological marker for the recognition and diagnosis of bipolar disorder.3.p-Ser9-GSK-3β/total GSK-3β in peripheral blood of bipolar mania patients are correlated with the severity of disease. |
