Objective: To research the expression of alpha B-crystallin(CRYAB)in gastric cancer tissues and explore the expression of CRYAB mRNA after neoadjuvant chemotherapy,analysis the relationship between CRYAB and characteristic of gastric cancer,and to explore its role and clinical significance in the occurrence and development of gastric cancer,as well as the effect of chemotherapy drugs on the expression of CRYAB mRNA.Methods: 1、The gastric cancer tissues and corresponding adjacent tissues of 76 patients underwent radical resection from April 2018 to March 2020 in Sichuan Mianyang 404 Hospital were collectedthe expression of CRYAB protein in the gastric cancer and normal gastric tissue of distal carcinoma of 76 patients with gastric cancer were detected by immunohistochemistry SP technique.The inclusion and exclusion criteria of gastric cancer patients were established according to the relevant literature.Inclusion criteria:(1)Age ≤ 75 years old,gastric cancer was diagnosed by preoperative endoscopic pathology,radical gastrectomy was performed according to the lesion site,postoperative pathological examination showed complete resection margin,and the resection margin was negative;(2)No anti-tumor treatment measures such as preoperative and intraoperative chemotherapy,preoperative radiotherapy,immunotherapy were taken;(3)All patients were diagnosed as primary gastric cancer by postoperative pathology,and the pathological type was confirmed;(4)The patients were diagnosed as primary gastric cancer(5)Paraffin specimens with complete tumor tissue and negative margin tissue.Exclusion criteria:(1)Patients with early gastric cancer undergoing endoscopic treatment [endoscopic mucosal resection(EMR)or endoscopic submucosal dissection(ESD)];(2)Patients with postoperative recurrence of gastric cancer or recurrent cancer of remnant stomach.The clinicopathological data of the patients were collected,including age,gender,tumor location,tumor diameter,TNM stage,Borrmann classification,degree of differentiation,lymph node metastasis,depth of invasion and pathological Lauren classification.The relation between the expression of CRYAB protein and clinicopathologic features was analyzed.2、 Twenty-one gastric tissues of patients accepted neoadjuvant chemotherapy and 26 gastric tissues of patients with no neoadjuvant chemotherapy in Sichuan Mianyang 404 Hospital were collected from November 2018 to March 2020,The inclusion and exclusion criteria of gastric cancer patients were established according to the relevant literature.Inclusion criteria:(1)Age ≤ 75 years old,gastric cancer was diagnosed by preoperative endoscopic pathology,radical gastrectomy was performed according to the lesion site,postoperative pathological examination showed complete resection margin,and the resection margin was negative;(2)No other antitumor therapy was performed except experimental chemotherapeutic drugs;(3)All patients were diagnosed as primary gastric cancer by postoperative pathology,and the pathological type was confirmed;(4)The clinical stage of all cases was Ⅱ-Ⅲ;(5)There was no significant underlying disease and he was able to tolerate surgery and postoperative clinical treatment.Exclusion criteria:(1)Patients with early gastric cancer undergoing endoscopic treatment [endoscopic mucosal resection(EMR)or endoscopic submucosal dissection(ESD)];(2)Patients with postoperative recurrence of gastric cancer or recurrent cancer of remnant stomach.(3)Patients with advanced lymph node metastasis.the expression of CRYAB mRNA was detected by rear time-PCR..Results:The expression of CRYAB protein in gastric cancer tissues was positive in 51 cases(67.11%)and the corresponding adjacent tissues in 32 cases(42.10%),the positive rate was higher in gastric cancer tissues(χ2=9.581,P=0.002).In the analysis of clinicopathological characteristics of gastric cancer tissues,the high expression of CRYAB protein in gastric cancer:In TNM stage,the expression of CRYAB protein in stage Ⅲ+ Ⅳ was higher than that in stage Ⅰ+ Ⅱ(P < 0.05);In histological differentiation degree of gasteic cancer,Low differentiation and undifferentiated group was higher than that in high differentiation and Medium differentiation group(P < 0.05);In invasion depth,the expression of CRYAB protein in plasma membrane layer and whole gastric layer was higher than that in mucosa layer and muscularis propria(P < 0.05);In Borrmann classification,the expression of CRYAB protein in and was Ⅲ Ⅲhigher than that in ⅠandⅡ group(P < 0.05)In Lauren’s classification,diffuse type was higher than intestinal type(P < 0.05),but had no correlation with age,gender,tumor location and tumor diameter(P > 0.05).The expression of CRYAB mRNA in the gastric cancer tissues with neoadjuvant chemotherapy was significantly higher than that in the gastric cancer tissues with no neoadjuvant chemotherapy(t=8.37,P<0.001)..Conclusion: 1.CRYAB protein is closely associated with the occurrence of gastric cancer.2.The expression of CRYAB protein in gastric cancer tissues is mainly related to the cell characteristics of gastric cancer cells,such as stage,type and degree of differentiation,but not related to general conditions,such as age and sex.3.The expression level of CRYAB protein is correlated with the malignant degree of gastric cancer to a certain extent,and plays a crucial role in the progression of gastric cancer.4.CRYAB mRNA transcription can be enhanced in gastric cancer tissues after chemotherapeutic drug stimulation. |
