| Aims:in order to further study and supervising g spirit on the treatment effect and its related mechanism of IBD,therefore,this experiment adopts the dextran sodium sulfate(DSS)induced by acute IBD mouse models to study collapse can g spirit by adjusting the ratio of Th17/Treg cells in mice and its related inflammatory factor levels,regulate the body’s environment,alleviate or treatment of inflammatory bowel disease(IBD),and explore its ease or treatment mechanism of IBD is associated with regulating the immune balance.Methods:Fifty 7-week-old C57BL/6J male mice were selected and in accordance with the principles of randomized,it is divided into 5 groups:Normal group,DSS control group,KKL low-dose group,KKL medium-dose group and KKL high-dose group.with 10 mice per group.The normal group was fed with free drinking sterile water for 8 days,and the DSS control group,KKL low-dose group,KKL medium-dose group and KKL high-dose group were given 4%DSS solution for 8 days to replicate the acute IBD mouse model.According to the equivalent drug dose of human and animal,sterile water and different doses of Chinese medicine were given to mice in each group on the second day of modeling,and intragastric operation was performed at the same time every day for consecutive 7 days.During the period of intragastric administration,the mental state,hair luster,diet and fecal characteristics of mice were observed,and the changes of body weight were weighed and recorded,and the disease activity index(DAI)score was made.On day 9,isoflurane anesthetized mice,serum samples were collected to detect the levels of serum IL-10 and IL-17A by ELISA.After the mice were killed,spleen tissue was taken and weighed to calculate spleen index.Flow cytometry was used to detect the ratio of Th17 and Treg cells in mouse spleen.The colonic length of each group was observed and measured by intercepting the part from anus to ileocecal colorectum.He staining was performed on the 1cm length of the colon 2cm above the anus of the mice.Images were collected under the microscope for pathology analysis and pathological scores were made.Results:compared with normal mice,the DSS in the control group mice on the third day appear listlessness,waxing loose,the symptom such as weight loss,DAI higher score and spleen index,colon length is shortened,the microscopic pathological visible incomplete colonic tissue structure,the mucous membrane of the damaged and destroyed gland,pathological assessment increases,its suppression of inflammation factor IL-10 levels drop,IL-17A proinflammatory factor levels rise,higher ratio of Th17/Treg,these results suggest acute IBD mouse model building success.Different doses of KKL(0.45g/mL,0.93g/mL,1.86g/mL)could slow down the weight loss of IBD mice,reduce DAI score and spleen index,inhibit the colonic shortened,protect the colonic tissue structure and the integrity of the colonic mucosa and glands to different degrees,relieve the inflammatory state of the mouse colon,increase the level of IL-10,inhibit the level of IL-17A,and reduce the Th17/Treg ratio,and the differences were statistically significant.Conclusions:Compared with the normal group,the proportion of Th17/Treg cells in the spleen of mice in the DSS control group was unbalanced and the levels of related inflammatory cytokines were abnormal,while KKL could alleviate the occurrence and development of IBD in mice in the DSS control group to a certain extent,and regulate the proportion of Th17/Treg cells in the spleen and the levels of related inflammatory cytokines.KKL can alleviate or cure IBD,which may be related to the regulation of the immune balance.It provides a more sufficient experimental basis for the clinical treatment of IBD with combined medicine of traditional Chinese and western medicine. |
