| OBJECTIVE:By detecting the number of Treg and Thl7 cells in the mesenteric lymph nodes of UC mice,the content of IL-10 and IL-17A in peripheral blood,and the expression of MPO,TNF-a and CP in colon tissue,the regulation of Teg and Th17 was studied.The mechanism of action of cells and related factors in the treatment of ulcerative colitis.MFETHODS:The UC mouse model was induced by 3.5%DSS solution and randomly divided into normal group,model group,5-ASA group and low-and high-dose group of ulceration enema,with 10 rats in each group.Body weight,stool texture and bleed ing were observed in each group,and the disease activity index was calculated.After the mice were sacrificed,the colon length was recorded and the intestinal weight index was calculated.The pathological sections of colon tissue were observed by light microscopy,and the histopathological changes of colon tissue were evaluated.The number of Treg and Th17 cells in mouse mesenteric lymph nodes was detected by flow cytometry.The levels of MPO,TNF-a and CP in serum and the levels of serum IL-10 and IL-1 7A in mouse colon tissues were determined by enzyme-linked immunosorbent assay.RESULTS:After modeling with DSS,the mice generally deteriorated,the DAI score increased significantly(P<0.01),and the colon tissue pathological condition was severe.Compared with the normal group,the colon length of the model group was significantly shorter(P<0.01),and the intestinal weight index was significantly increased(P<0.01).The number of CD4+CD25+Foxp3+Treg cells in the mesenteric lymph nodes was decreased(P<0.01),CD3+CD4+IL-The number of 17A+Th17 cells increased(P<0.01);the level of IL-10 in serum was down-regulated,and the level of IL-17A was up-regulated(P<0.01).The contents of MPO,TNF-a and CP in colon tissue were significantly increased.The difference was statistically significant.(P<0.05,P<0.05,P<0.01).After administration,compared with the model group,the weight loss rate of the 5-ASA group and the KGP low-dose group decreased(P<0.01,P<0.05,P<0.05),the general condition improved,and the DAI score decreased.P<0.05);the colon length of the three groups also increased,the intestinal weight index decreased significantly(P<0.05);the pathological condition of the colon tissue improved.The number of CD4+CD25+Foxp3+Treg cells in the mesenteric lymph nodes was significantly increased in the 5-ASA group and the KGP high-dose group(P<0.01),but the KGP low-dose group was increased but the effect was slightly lower than the former two(P<0.05).The number of CD4+ IL-17A+ Th 17 cells was significantly decreased(P<0.01).The levels of IL-10 in 5-ASA group and KGP low-dose group were up-regulated and IL-17A levels were down-regulated(P<0.01).The contents of MPO and TNF-a in colon tissue were significantly decreased(P<0.05),especially CP decreased(P<0.01).Conclusions:Kuijie Guanchang Prescription may regulate the immune balance of Treg/Th17 in UC mice,increase the number of Treg cells,up-regulate IL-10 levels,decrease the number of Thl7 cells,down-regulate the expression of IL-17A,MPO,TNF-a,and CP,and treat UC. |
