Effect Of Siyin Recipe On NLRP3 In MCAO Rats And Observation Of Clinical Efficacy

Goal:To explore the effect of NLRP3 on ischemia-reperfusion model rats and the effect of Siyin Recipe on patients with cerebral infarction at the recovery stage(phlegm and blood stasis obstructing the collaterals).Method:Experimental study: 54 rats were randomly divided into 3 groups,namely the sham operation group,the model group,and the four-factor prescription group.Neurological function scores were performed on the 1,7,and 14 days after cerebral ischemia and reperfusion,and the survival rate of the mice was compared.Western blotting was used to examine the expression of NLRP3 in the hippocampus on the ischemic side of each group at each time;Clinical study: A total of 67 patients were randomized and completed observation.The treatment group(34cases)received Siyinfang combined with conventional Western medicine treatment,and the control group(33 cases)received conventional Western medicine treatment.Before and after treatment,the two groups were given TCM syndrome score,NIHISS score,and modified Rankin score to compare their differences.Experimental research results:1.Survival curve comparison,the survival curve of the model group is lower than that of the sham operation group(P<0.05),and the survival curve of the four factors group is higher than that of the model group(P<0.05).Siyinfang can improve the survival rate of MCAO model rats.2.Neural function scores were compared in pairs,and the scores of the sham operation group(1 day,7 days,14 days)were all 0 points.The neurological function scores of the model group increased after modeling(1 day,7 days,14 days),compared with(at the same time point)the sham operation group(P<0.05).The scores of neurological function in the Siyinfang group on 7 and 14 days were lower than that of the model group(P<0.05),which indicated that Siyinfang could improve the neurological deficit symptoms in rats with cerebral ischemia.3.Compared with the expression of NLRP3 protein,the NLRP3 protein in the hippocampus on the ischemic side of the model group was higher than that in the sham operation group on day 1,7 and 14(P<0.05),indicating that cerebral ischemia can increase the expression of NLRP3 protein in rats;The NLRP3 protein in the ischemic hippocampus of the rats in the 1st,7th,and 14 th days was lower than that of the model group(P<0.05),suggesting that Siyinfang can down-regulate the expression of NLRP3 protein in the hippocampus of the ischemic side of rats.Clinical research results:1.After treatment,the total effective rate was 88.24% in the treatment group and72.73% in the control group.There was a difference between the two groups(P<0.05),and the treatment group was better than the control group.2.After treatment,there were differences between the two groups in the curative effect of astringent language,numb limbs,pale complexion,excessive or sticky phlegm,dry stool,constipation,insomnia and dreaminess,shortness of breath and fatigue(P<0.05).The treatment group was better than the control group.There was no significant difference in the efficacy of "hemiplegia,crooked mouth and tongue,dizziness,and choking when drinking water"(P>0.05).3.The NIHSS scores between the two groups after treatment were different(P<0.05),and the treatment group was lower than the control group.The modified Rankin score between the two groups after treatment showed no significant difference in scores(P>0.05).4.After treatment,the scores of the two groups of patients were lower than that before treatment(P<0.05),indicating that the two treatment methods are effective for the evaluated symptoms.Completion:1.Siyin Recipe can improve the neurological deficit symptoms of MCAO reperfusion model rats,increase the survival rate of the rats,and reduce the expression of NLRP3 related protein in the hippocampus of the ischemic side,and reduce the inflammatory response in the rats.2.Siyin Recipe can reduce the neurological deficit score and the modified Rankin score of patients with cerebral infarction in the recovery period(phlegm and blood stagnation syndrome)."Insomnia,dreaminess,shortness of breath,fatigue" and other symptoms are better than the control group,with good curative effect and relatively safe.
