Objective: In this study,the mouse model of cerebral hemorrhage was used as the research object.The neurological function score,brain water content,he staining,Western blot detection,immunohistochemistry and other methods were used to study whether the traditional Chinese medicine decoction of activating blood and removing stasis and supplementing essence and marrow could improve the degree of nerve injury and inhibit cell apoptosis in mice with cerebral hemorrhage by regulating Nrf2.Methods: 78 normal wild mice and Nrf2 knockout mice were randomly divided into three groups: sham operation group,model group and prescription group,26 in each group.The model of cerebral hemorrhage was made by injecting the tail artery blood into the brain of the mice by using rynkowski model.After the model was successfully established,Garcia was used to evaluate the neurological function defects of each group of models.The brain water content was measured by dry and wet weight method.The pathological changes of brain tissue in mice were observed by HE staining method.The intervention effect of traditional Chinese medicine decoction was studied.The expression of Caspase-3,Bax and Bcl-2 in the brain of mice was detected by Western blot and immunohistochemistry.The effects of traditional Chinese medicine decoction of activating blood and removing stasis and supplementing essence and marrow on apoptosis of brain cells were studied.Results:1.The Garcia neurobehavioral score of mice in formula groupwas higher than that in model group(P < 0.05),and there was significant difference between wild group and gene knockout group(P < 0.05).2.Compared with sham operation group,the brain water content of model group increased significantly(P < 0.05),and that of formula group decreased significantly(P <0.05).3.The pathological changes of brain tissue in model group and formula group were obvious by HE staining,but the damage of brain tissue in formula group was lighter than that in model group,and there was significant difference between wild group and gene knockout group(P < 0.05).4.Western blot was used to detect the expression of Caspase-3,Bcl-2 and Bax.The expression of Caspase-3 and Bax in model group and formula group was higher than that in sham operation group(P < 0.01),and the expression of Bcl-2 was lower than that in sham operation group;Caspase-3 and Bax in the formula group were lower than those in the model group(P < 0.05),and Bcl-2 was higher than that in the model group(P < 0.05).There was no significant difference in Caspase-3 but significant difference in Bcl-2 and Bax between the two groups(P < 0.05).Caspase-3 and Bax in the formula group were lower than those in the gene knockout formula group(P < 0.05),and Bcl-2 in the formula group was higher than that in the gene knockout formula group(P < 0.05).5.The expression of Caspase-3 and Bax in the model group was higher than that in the sham operation group.The levels of Caspase-3 and Bax in the prescription group were lower than that in the model group(P < 0.05),and Bcl-2 was higher than that in the model group(P < 0.05).The expression of Caspase-3 and Bax protein in the prescription group was lower than that in the group of gene knockout prescription(P < 0.05),and Bcl-2 was higher than that in the group of gene knockout prescription(P < 0.05).DAB was mainly stained,while the color of apoptosis cells in model group and prescription group was yellow.The staining of the cells in the prescription group was obviously lighter.Conclusion:1.he traditional Chinese medicine decoction of activating blood and removing stasis and supplementing essence and marrow can effectively reduce the degree of brain edema after intracerebral hemorrhage,reduce the inflammatory reaction of brain tissue around hematoma,and improve the degree of neurological deficit.2.The inhibition of brain hemorrhage injury by activating blood and removing stasis and supplementing essence and marrow decoction is achieved by reducing the expression of Caspase-3 and Bax,and improving the expression of bcl-2.3.The traditional Chinese medicine decoction of activating blood and removing stasis and supplementing essence and marroww can reduce the secondary injury of cerebral hemorrhage by regulating Nrf2.
