Objective:Explore the therapeutic effect of Yuye Decoction on rats with type 2diabetic nephropathy,observe and compare the body weight,blood glucose,renal function,renal pathomorphological changes,and the expression of HMGB1/TLR4 Protein in renal tissue,and explore its therapeutic mechanism and pathway.Methods:100 healthy SD male rats were selected,10 were randomly selected as the normal group,and they were given regular feeds.The remaining 90 were fed a high-fat and high-sugar diet for 6 weeks combined with Streptozotocin(STZ)to induce type 2 diabetic nephropathy rat model,60 rats meeting the model criteria were screened and randomly divided into four groups,namely the model group,the Yuyetang high-dose group(11.08g·kg-1),and the Yuyetang low-dose group(5.54g·kg-1)and metformin group(200mg·kg-1),15 rats in each group.The normal group and the model group were given an equal volume of physiological saline by gavage,once a day,and sacrificed after 8 weeks of continuous treatment.The weight,blood glucose,and renal function levels of the rats in each group were measured.The pathomorphological changes of kidney tissue were observed under an optical microscope.Detect the expression levels of HMGB1 protein and TLR4 protein in kidney tissue.Results:1.Compared with the model group,the blood glucose and body weight of the rats in the treatment group were improved.2.The pathological results showed that the glomerulus of the rats in the Yuyetang high-dose group was normal,and there was no ballooning of renal tubular epithelial cells.The kidneys of rats in the Yuyetang low-dose group and metformin group were better than those in the model group,but there were still local renal corpuscle enlargement,glomerular interstitial widening,balloon adhesion,and the cytoplasm of tubular epithelial cell was loosened,the degree of edema was reduced,and there was no necrosis of the renal tubules.3.Compared with the model group,the serum Urea,Scr,and UA levels in the treatment group decreased,which was different(P<0.05).4.Compared with the model group,the expression of HMGB1 protein and TLR4protein in the kidney tissue of the treatment group was significantly reduced(P<0.01),and the Yuyetang high-dose group had the best effect.Conclusion:1.Yuye Decoction in the treatment of type 2 diabetic nephropathy rats has the effect of improving glucose metabolism,body weight and renal function,and can delay the development of diabetic nephropathy.2.The mechanism of Yuye Decoction protecting the kidneys of type 2 diabetic nephropathy rats is related to reducing the level of HMGB1 protein,inhibiting the TLR4signaling pathway,and reducing the inflammatory response in kidney tissue.
