ObjectiveIn diabetes,the blood-brain barrier is damaged,harmful substances penetrate the blood-brain barrier and damage brain tissue,resulting in cognitive decline.Salidroside is a natural extract of salidroside,which has a wide range of biological functions.This study is to investigate the effects of salidroside on blood-brain barrier and cognitive function in diabetic rats,salidroside for the prevention and treatment of diabetic cognitive impairment to provide a theoretical basis.MethodsUsing a random number table method,healthy SD rats were divided into three groups: the control group,the diabetes group,and the salidroside treatment group.The intraperitoneal injection of streptozotocin was used to divide the diabetes group and the salidroside treatment group SD rats were prepared as a diabetes model,and the control group was injected intraperitoneally with sodium citrate buffer.After the model was successfully established,the SD rats in the salidroside treatment group were given Sal(50mg/kg/day)gavage treatment,the control group The SD rats in the group and the diabetes group were treated with an equal volume of normal saline by gavage.After 12 weeks,the body weight and blood glucose level of each group of rats were measured,and the learning ability and memory ability of each group of rats were measured by the water maze experiment,Evans Blue method was used to detect BBB permeability,Western blot method was used to detect the expression changes of tight junction proteins Occludin and ZO-1 in the hippocampus of each group of rats,and TUNEL staining was used to detect the apoptosis of neurons in the hippocampus of each group.ResultsCompared with the control group,the rats in diabetes group lost weight,increased blood glucose,decreased learning and memory ability(P < 0.05),increased blood-brain barrier permeability(P < 0.05),decreased the expression of ocudin and ZO-1(P < 0.05)。The number of TUNEL positive cells increased(P<0.05);Compared with the diabetic group,salidroside treatment group had no significant changes in body weight and blood glucose(P > 0.05),learning and memory ability was improved(P < 0.05),blood-brain barrier permeability was decreased(P < 0.05),and the expressions of Occludin and ZO-1 were increased(P < 0.05).The number of TUNEL positive cells decreased(P <0.05).ConclusionsSalidroside can promote the expression of tight junction protein,improve the function of blood-brain barrier,inhibit the apoptosis of hippocampal neurons,improve the learning and memory ability of diabetic rats,and prevent the cognitive impairment of diabetes. |
