ObjectiveSalidroside(SAL) can effectively alleviate the cognitive impairment of Alzheimer’s disease(AD),but its effect on the learning and memory ability of diabetic encephalopathy(DE)remains unclear.The aim of this study was to investigate whether salidroside could improve the learning and memory ability of diabetic rats by inhibiting neuronal pyroptosis in hippocampus.MethodsThirty healthy male Sprague Dawley rats were randomly divided into control group(control group),diabetes group(DM group)and salidroside group(SAL group),with 10 rats in each group.Diabetic rats and salidroside groups were induced by intraperitoneal injection of streptozotocin(STZ)(60mg/kg).The tail vein blood glucose of the rats was measured 3 days after STZ injection.If the blood glucose concentration was higher than 16.7mmol/L,the diabetic rats were successfully established.Rats in salidroside group were given salidroside 100mg/kg by gavage 7 days after successful modeling;rats in control group and diabetes group were given saline solution by gavage,once a day for 12 weeks.After 12 weeks,Morris water maze test was used to test the spatial learning and memory ability of rats.The expressions of NLRP3,caspase-1,gsdmd,IL-1βand IL-18 were detected by Western blot.ResultsCompared with the diabetic group,the blood glucose level of salidroside group was lower(18.9±2.1)mmol·L-1,and the body mass level was higher(242±25)g(P<0.05);in the water maze directional navigation experiment,the escape latency of control group,diabetic group and salidroside group were(10.0±2.1)s,(41.5±11.2)s,(23.8±9.3)s respectively.Compared with the control group,the escape latency of diabetic group was significantly longer(P<0.01);compared with the diabetic group,the escape latency of salidroside group was significantly shorter(P<0.05).In the water maze space exploration experiment,the crossing time of control group,diabetes group and salidroside group was(32.5±3.8)s,(18.6±6.9)s and(27.8±5.4)s respectively.Compar-ed with the control group,the time of diabetic rats in platform quadrant was significantly shorter(P<0.01);compared with the diabetic group,the time of salidroside group in platform quadrant was significantly longer(P<0.05);Western blot results showed that compared with the control group,the expression of NLRP3,caspase-1,gsdmd,IL-1βand IL-18 protein in the hippocampus of rats in the diabetic group was significantly increased(P<0.01);compared with the diabetic group,the expression of NLRP3,caspase-1,gsdmd,IL-1βand IL-18 protein in the hippocampus of rats in the salidroside group was decreased(P<0.01).ConclusionsSalidroside can improve the learning and memory ability of diabetic rats.The mechanism may be related to the inhibition of focal pyroptosis of neurons in the hippocampus of brain. |
