ObjectiveHuman tongue squamous cell carcinoma cell line Tca8113 was treated with Isomalactone in vitro.The effects of Isomalactone on the proliferation,migration and invasion of tongue squamous cell carcinoma cell line Tca8113 were studied by observing the phenotypic experiment,and its related mechanism was discussed.MethodsThe 50%inhibitory concentration(IC50)of Alternaria mandshurica lactone on tongue squamous cell carcinoma Tca8113 cells was detected by CCK-8 method,and the optimum concentration of Alternaria lactone solution on tongue squamous cell carcinoma Tca8113 cells was determined.Cologne formation test and EDU test were used to detect the effect of Aristolactone on the proliferation of Tca8113 cells.After fixed staining,the experimental results were observed under fluorescence microscope.The scratch test was used to examine the effect of iso-arginolide on the migration ability of Tca8113 cells.Transwell assay was used to examine the effect of iso-arginolide on the invasive ability of Tca8113 cells.The apoptosis of Tca8113 cells was detected by TUNEL apoptosis detection kit.Pass the expression of apoptosis-related proteins in Tca8113 cells was detected by Western blot method.ResultsThe 50%inhibitory concentration(IC50)of 20pM on tongue squamous cell carcinoma Tca8113 cells was 20pM by CCK-8 method,and it was co-cultured with tongue squamous cell carcinoma Tca8113 cells.The results showed that with the change of time,the cell activity decreased,and the Tca8113 cell activity decreased gradually(p<0.01).The results of colony formation assay and EDU assay showed that Alternaria lactone could inhibit the proliferation of Tca8113 cells,inhibit the migration of Tca8113 and decrease the invasive ability of Tca8113 cells(p<0 01).The staining results showed that Equisearia lactone could promote the apoptosis of Tca8113 cells(p<0 01).The results of Western blot assay showed that isoglycerolide could down-regulate the Bcl-2 protein and up-regulate the expression of caspase3 protein in tongue cancer Tca8113 cells,indicating that it can promote the apoptosis of Tca8113 cells.ConclusionsAlternaria lactone can inhibit the proliferation,migration and invasion of tongue squamous cell carcinoma Tca8113 cells and promote Tca8113 apoptosis in a concentration-and time-dependent manner.However,the specific mechanism is clear,so further experimental research is needed. |
