Effect Of Combined Treatment Of Hirudin With Adriamycin On Anti-proliferation Of Tongue Squamous Cell Carcinoma Line TCA8113

Objective :To investigatue the effect of hirudin combined with adriamycin(ADM)on anti-proliferation of tongue squamous cell carcinoma line TCA8113.Methods :The inhibitory effect on cell proliferation was assessed by MTT assay. The cell cycle of TCA8113 cells was detected with Flow Cytometry(FCM).Transmission electron microscopy and fluorescent staining were applied to demonstrate the presence of apoptosis. Results: Hirudin and ADM could inhibit TCA8113 cell proliferation and induce apoptosis in a time-dose dependent manner(P<0.05).The combination of Hirudin(15ATU/ml) and ADM(0.39-3.125ug/ml) showed enhanced joint killing effect(P<0.05). The blocking of the cell cycle was in G0/G1 -phase .Apoptotic feature was observed by transmission electronmicroscopy and fluorescent staining.Conclusion :Hirudin could significantly inhibit the proliferation of TCA8113 cells . The mechanism might be inducing apoptosis and affecting the distribution of cell cycle.
