The Establishment Of The Method Of The Multi-components Quantities By One Marker For The Total Flavonoids Effective Fraction Of Ziziphora Clinopodioides Lam.and A Preliminary Study Of Its Antiatherogenic Activity

Objective: to establish a method of the Multi-components Quantities by One Marker of for the total flavonoids effective fraction of Ziziphora clinopodioides Lam.,to preliminarily investigate its antioxidant and antiatherogenic activities,so as to establish the quality standard for the effective sites of the total flavonoids effective fraction of Ziziphora clinopodioides Lam.,and to provide a reliable basis for further research and rational utilization of resources.Methods: 1)HPLC method was used to analyze the chemical composition of total flavones of Ziziphora clinopodioides Lam;2)ABTS+· free radical,·OH free radical,superoxide anion(O2-·)free radical,DPPH free radical and FRAP were used to evaluate the antioxidant activity of total flavones of Ziziphora clinopodioides Lam.in vitro;3)cultivate people THP-1 cells,mononuclear cell line with PMA differentiation as macrophages,ox-LDL induced THP-1 macrophages to foam cell,through the adoption of CCK-8 method to detect the cell vitality,oil red O staining observation of lipids in the cell,ELISA to detect the contents of TC,FC,TG in foam cells,determination of FC/TC value inside the cell,evaluation of Ziziphora clinopodioides Lam.flavonoids in vitro activity of atherosclerosis.Results: 1)HPLC method was established to compare the total flavonoids effective fraction of Ziziphora clinopodioides Lam.with the standard reference;2)a HPLC method was established for the simultaneous determination of various flavonoids in total flavones of Ziziphora clinopodioides Lam;3)a method of the Multi-components Quantities by One Marker of total flavones in Ziziphora clinopodioides Lam.was established;4)the results of in vitro antioxidant experiments showed that the total flavones of Ziziphora clinopodioides Lam.had a strong scavenging ability on ABTS+· free radical,·OH free radical,superoxide anion(O2-·)free radical and DPPH free radical.The IC50 values were 24.76μg/m L,0.55mg/m L,0.06mg/m Land 0.34mg/m L,respectively.The total flavones of Ziziphora clinopodioides Lam.have strong antioxidant capacity and concentration dependence;5)Total flavones of Ziziphora clinopodioides Lam.have antiatheromatous activity.Total flavones and monomer linarin can inhibit the formation of foam cells,reduce the accumulation of lipid components in plaque,and have anti-atherosclerosis activity to some extent.Conclusion:In this paper,a HPLC method and the Multi-components Quantities by One Marker method were established for the determination of various flavones in the total flavones of Ziziphora clinopodioides Lam.,proving that the total flavones of Ziziphora clinopodioides Lam.have strong antioxidant activity and anti-atherosclerosis activity,thus for Ziziphora clinopodioides Lam.effective parts of flavonoid in quality standards,for its further research and rational utilization of resources provides a reliable experimental and theoretical basis.
