| Objective: To study the extraction and purification of flavonoids ingredients fromUygur medicine Ziziphora clinopodioides Lam., and the development ofimmediate-release formulations in the treatment of cardiovascular drugs-dropping pills,lay the foundation for its mechanism research and new drug development. Methods:Selected the content of total flavonoids as an indicator, and to choose the best extractionprocess and inspect the purification performance of ten kinds of macroporous resins,preferred a resin and a process of enrichment of total flavonoids. To analyse flavonoidsingredients by LC-MS/MS. Used the orthogonal design to optimize technology ofdropping pills. The removing DPPH· method for the determination of total flavonoids freeradical scavenging. Results: The best extraction process were:25times of70%alcohol,refluxing and extracting for3time, each time1.5h. The best resin was XDA-1, the bestpurification process were: concentration of extract2.515mg/ml; absorption flow rate3BV/h;30%ã€50%ã€70%alcohol as the eluant; the column was washed with30%alcoholã€50%alcohol4BV and70%alcohol3BV,respectively. The analysis of flavonoidsingredients by LC-MS/MS were: hyperoside, chrysin, genkwanin,5,6,4’-trihydroxy-7,8,3’-trimethoxy flavones and acacetin. The best moulding technical condition: the refrigerantwas dimethylsilicon oil, the temperature of refrigerant was10~15℃; the scale ofmedicine:PEG6000:PEG4000were1:1:1; the temperature of drugs was80℃. Theefficacy studies of ZCF have shown: there was a good Dose-effect relationship betweenZCF concentration and DPPH radical clearance rate. IC50of ZCF was0.017mg/ml. ZCFcan significantly decrease MDA content in the model of hypoxia/reoxygenation injury (P﹤0.05). Conclusion: The extraction process, simple and highly reproducible.Optimization of the XDA-1macroporous resin can be effectively enriched Ziziphoraclinopodioides Lam.total flavonoids.By LC-MS/MS technology to determine the activeingredients of Ziziphora clinopodioides Lam. flavonoids. The preparationprocess of the drop pills in linewith the Pharmacopoeia of the People’s Republic of China. ZCF has apotential antioxidant activity and protective effects on neonatal cardiomyocytes damageinduced by hypoxia/reoxygenation stress through reduction of lipid peroxidation andenhancement of the activity of antioxidant defense. |
