Establishment Of The Diagnostic Scale Of Syndrome Of The Heart Failure Qi-Yin Deficiency And The Study On The Effect Of The Expression Of Calcium Regulatory Protein In Heart Failure Rats By Astragalus Membranaceus、Monkshood And Lepidium Seed

Objective:To establish the diagnostic scale of syndrome of the Chronic Heart Failure Qi-Yin deficiency,and to provide theoretical basis for standardization and objectivity of TCM syndrome quantitative diagnosis of the chronic heart failure with Qi-Yin deficiency;To explore the effect of Astragalus membranaceus、Monkshood and Lepidium seed on the expression of calcium regulatory protein in cardiac myocytes of rats with heart failure,and to study the mechanism of its treatment of chronic heart failure.Methods:1.Establishment of the diagnostic scale of syndrome of the Chronic Heart Failure Qi-Yin deficiencyFirstly,after the ancient and modern literature retrieval and Research on the Qi-Yin deficiency in chronic heart failure,expert questionnaires were made and investigated to form the first edition of the diagnosis scale of the chronic heart failure Qi-Yin deficiency.Secondly,the second edition of the diagnostic scale was formed by using a variety of statistical methods to carry out scale tests and item selection,and the accuracy of the scale was evaluated from the aspects of reliability、validity、responsiveness and so on.Finally,on the basis of clinical epidemiological survey,the scale is used as a measurement tool to calculate the index weight coefficients.2.the Study on the effect of the expression of calcium regulatory protein in heart failure rats by Astragalus membranaceus、Monkshood and Lepidium seedSixty rats were randomly divided into six groups:blank group,model group,astragaloside group,trimetazidine group,low dose group of Chinese herbs and high dose group of Chinese herbs,Adriamycin hydrochloride was injected into abdominal cavity to establish heart failure model,and astragaloside,trimetazidine hydrochloride,low dose and high dose of Chinese herbs were used for drug intervention,at the 4th and 8th week of the experiment.echocardiography and plasma B-type brain natriuretic peptide(BNP)were measured.At the 8th week of the experiment,the left ventricular tissue of the rats was taken,Western blot and RT-PCR were used to detect the expression of NCX1 and SERCA2 in cardiac sarcoplasmic reticulum of rats with heart failure,To explore the effect of Astragalus membranaceus、Monkshood and Lepidium seed on the expression of NCX1 and SERCA2 in cardiac myocytes of rats with heart failure.Results1.Establishment of the diagnostic scale of syndrome of the Chronic Heart Failure Qi-Yin deficiencyThis study established the diagnosis scale of the Chronic Heart Failure Qi-Yin deficiency.After comprehensive evaluation,the scale has good reliability and validity.On this basis,the diagnostic model and criteria for the Qi-Yin deficiency were established.The diagnostic threshold was 305.5.2.the Study on the effect of the expression of calcium regulatory protein in heart failure rats by Astragalus membranaceus、Monkshood and Lepidium seed RT-PCR showed that the expression levels of NCX1 and SERCA2 were different in different groups,the expression level of high dose group and low dose group was significantly higher than other groups.Western blot showed that at the protein expression level of NCX1 and SERCA2,there was no significant difference between low dose group,high dose group and other groups.Conclusion:1.Establishment of the diagnostic scale of syndrome of the Chronic Heart Failure Qi-Yin deficiencyThe diagnostic scales of the Chronic Heart Failure Qi-Yin deficiency syndrome established by literature research,clinical questionnaires and statistical analysis have good representativeness,feasibility and practical value.The research results are the syndrome score of the threshold of the diagnosis of Qi-Yin deficiency,Y≥305.5,that is,when the syndrome score of patients with Chronic Heart Failure is Y≥305.5,the diagnosis of Qiyin deficiency syndrome is established.2.The experimental results showed that Astragalus membranaceus、Monkshood and Lepidium seed could improve the heart function of heart failure rats and reduce the plasma BNP level by inhibiting the expression of NCX1 and increase the expression of SERCA2,so as to realize the treatment of chronic heart failure.
