Objective:MicroRNA is an endogenous non-coding single-stranded small RNA molecule that plays an important role in gene regulation during the occurrence of polycystic ovary syndrome(PCOS).Based on the characteristic expression of microRNA,this study was used to observe the expression changes of basic endocrine and microRNA-183/200/223 expression changes.In this study,based on the characteristic expression of microRNA,the patients with PCOS phlegm and dampness syndrome were observed to observe the basic endocrine and microRNA-183/200/223 expression changes in serum,follicular fluid,and follicular granulosa cells under the treatment of dampness-drying and phlegm-resolving prescription Qi Gong Wan.To explore the mechanism of the dampness-drying and phlegm-resolving prescription on PCOS phlegm-dampness syndrome,and to provide new ideas and theoretical basis for the study of traditional Chinese medicine in the treatment of PCOS phlegm-dampness syndrome.Method:A total of 40 PCOS patients with phlegm dampness who were treated with IVF assisted pregnancy were selected for treatment in the affiliated hospital of Shandong University of Traditional Chinese Medicine(randomly divided into treatment group and control group,20 cases in each group).The height and weight of the patients in each group were measured,and the body mass index(BMI)was calculated.The chemiluminescence method was used to detect the sex hormone levels in each group.Quantitative real time polymerase chain reaction(qRT-PCR)was used to detect the expression levels of miR-183/200/223 in each group of serum.The treatment group was treated with Qi Gong Wan adjuvant therapy for two months and the above indicators were tested again.Both groups(the treatment group was two months after Qi Gong Wan adjuvant treatment)used the gonadotropin-releasing hormone agonist(GnRH-a)/follicle-stimulating hormone(FSH)long protocol to promote ovulation and B-ultrasound guided transovarian puncture to retrieve eggs.qRT-PCR was used to detect the expression levels of miR-183/200/223 in follicular fluid and granulosa cells of the two groups of patients.Observe the changes in TCM syndrome scores,basal endocrine,and changes in miR-183/200/223 in serum,follicular fluid,and granulosa cells in the treatment group before and after Chinese medicine intervention,and between the treatment group and the control group.Results:1.General information and basic endocrine comparison:there were no statistically significant differences in the age,BMI,and basic endocrine indexes between the treatment group and the control group before the intervention of traditional Chinese medicine;after intervention,the LH value,T value and LH/FSH value decreased significantly,and the differences were statistically significant(P<0.01).2.Comparison of TCM Syndrome Points:there was no statistical difference between the treatment group and the control group before the intervention of Chinese medicine,and the score decreased after the intervention,the difference was statistically significant(P<0.01).3.Comparison of microRNAs in serum:there was no statistical difference between the treatment group and the control group before the intervention of Chinese medicine;the relative expression of miR-223-3p decreased after the intervention,the difference was statistically significant(P<0.05),the relative expression of miR-200c-3p decreased,but the difference was not statistically significant.4.Comparison of microRNA in follicular fluid:the relative expression of miR-200c-3p increased and the relative expression of miR-223-3p decreased in the treatment group,and the differences were statistically significant(P<0.01).5.Comparison of microRNAs in granule cells:the relative expression of miR-200b-3p and miR-223-5p increased in the treatment group,while the relative expression of miR-200c-3p and miR-223-3p decreased,the differences were statistically significant(P<0.01);and the relative expression of miR-183-5p increased,but the difference was not statistically significantConclusion:Qi Gong Wan decoction for dryness and phlegm can effectively improve the TCM syndromes of patients with PCOS phlegm-dampness syndrome,reduce the serum LH value,T value and LH/FSH value of patients,and correct the abnormal expression of miR-183/200/223 in patients.So as to treat PCOS phlegm dampness.
