| Ganhuangcao,whose scientific name is Penthorum chinense Pursh,is the only one plant of the Penthorum genus.As a traditional medicine of the miao nationality,it has the functions of clearing away heat,detoxifying,removing jaundice,and clearing damp.Wild Penthorum chinense is widely distributed in China and the countries of southeast Asian,but only the plant growing in China can be used as medicine.It has been domesticated and planted on a large scale,and only the product in Sichuan is authentic medicinal materials.Gansu granule,which is produced from the single herb of Penthorum chinense,has been proved to have good liver protecting activity in clinical practice,but its active ingredients and pharmacological mechanism are not clear at present.Therefore,this experiment starts from these two aspects,in order to provide the basis and research direction for the further study of Penthorum chinense.HPLC-QTOF/MS(LC-MS)was used to establish a detection method of the chemical constituents of Penthorum chinense and the methodology was verified.On this basis,the fingerprint of ethanol extract of Penthorum chinense was established.Through the analysis of the primary and secondary mass spectrometry data,as well as the retrieval,speculation and identification of the chromatographic peaks by CNKI,sicfinder and other databases,32 compounds were obtained.Through principal component analysis and cluster analysis,the quality differences among 10batches of Penthorum chinense produced by different manufacturers were investigated(Y1:Gulin Ganhuangcao cooperative in Sichuan;Y2:Kangjia pharmaceutical co.LTD.in Chongqing;Y3:Gulin Hongan pharmaceutical co.,LTD.in Sichun;Y4:Shuangcheng medicine co.LTD.in Sichuan;Y5:Qiaoyi Baicao in Sichuan;Y6:Huachunliang;Y7:West Jue;Y8:hundred years and food;Y9:Zangxi hall;Y10:Deng’s traditional Chinese medicine shop in Sichuan).The results showed that except for samples Y6 and Y9,the quality of the other eight batches of samples was basically the same,and the comprehensive score of sample Y10 was the highest,which laid the foundation for the further improving of the quality and deep development of Penthorum chinense.Through the network pharmacology research,the main active ingredints of Penthorum chinense for the treatment of liver fibrosis were screened,the targets of their active ingredients were predicted,a"drug component-target-disease"network was established,and the mechanism of Penthorum chinense for treating liver fibrosis was explored.A total of 189 targets related to 22 chemical constituents,949 targets related to liver fibrosis disease,and 82 targets intersecting of the two ways were found through the databases.The protein interaction network was buildt up through the String database and Cytoscape 3.7.2 software,and the results showed that SRC,PIK3R1,PIK3CA are three key targets.The topological analysis results of the"drug component-target-disease"network showed that Kaempferol,Ganhuangcaosu F,and Ganhuangcaotong C may be the potential active ingredients.And through the KEGG pathway analysis,the possible signal pathway of Penthorum chinense for the treatment of liver fibrosis were predicted.The results of network pharmacology provided a research direction and theoretical basis for the further study of the anti-fibrotic active ingredients and the mechanism of action of Penthorum chinense.Acute liver injury induced by CCl4was used to establish a liver injury model in rats,and the intervention and influence of the 70%ethanol extract of Penthorum chinense on liver fibrosis during liver injury repair were investigated.The results showed that the 70%ethanol extract of Penthorum chinense can sufficiently reduce the activity of ALT in the plasma of liver injury rats,and the results of western blot showed that this extract can significantly reduce the expression of TNF-αand TGF-βin liver tissues,indicating that Penthorum chinense can interfere with and affect the liver fibrosis process in the repair of acute liver injury induced by CCl4,and its mechanism may be related to the inhibition of inflammation. |
