Objective:Astaxanthin is a carotenoid with the potential oxidation resistance and antitumor activity.However,it has not been reported that the interaction between astaxanthin and cervical cancer cells at home and abroad.In this study,cervical cancer cell lines(HeLa and CaSki)were used as experimental research objects,and the effects of astaxanthin with different concentrations on the biological behavior of cervical cancer cells were investigated.The aim is to provide experimental basis for the further application of astaxanthin.Methods: The effects of astaxanthin on the proliferation of cervical cancer cells were detected by MTT and trypan blue staining assay.The effects of astaxanthin on the cell migration and invasion of cervical cancer were studied by cell scratch and Transwell invasion experiments.The effects of astaxanthin on apoptosis of cervical cancer cells were detected by Hoechest33258 nuclear staining and flow cytometry.Western blot assay was used to detect the expression of apoptotic proteins(PARP and Caspase3)and the proteins related to the MAPK/JNK signaling pathway(p-JNK,p-ERK and p-P38).Results: The cells of HeLa and CaSki were significantly inhibited by treated with astaxanthin at 100-300 M,which exhibited a dose-dependent manner.The results of cell scratch and Transwell assay indicated that the astaxanthin can vividly inhibit the migration and invasion of cervical cancer cells.In the Hoechest33258 staining experiment,abnormal nuclear shrinkage and fragmentation induced by astaxanthin,and the generated apoptotic bodies were observed by fluorescence microscope.Flow cytometry showed that astaxanthin can significantly increase the number of cervical cancer cell apoptosis.The results of western blot showed that the astaxanthin could promote the expression of apoptotic proteins(PARP,Caspase3)and the related proteins in the JNK signaling pathway.Nevertheless,the JNK preparation(SP600125)can result in the promoting effect of astaxanthin on Caspase3 and PARP in cervical cancer cells distinctly reduced.Conclusion: Astaxanthin has an obvious influence on the biological behavior of cervical cancer cell lines.It can inhibit the proliferation,migration and invasion of cervical cancer cells and induce apoptosis by activating the JNK signaling pathway of cervical cancer cells.
